大肠癌组织中VEGF-C、STAT3的表达变化及意义

     

摘要

目的 观察大肠癌组织中血管内皮生长因子C(VEGF-C)、信号转导和转录激活因子3(STAT3) mRNA及其蛋白的表达变化,并探讨其临床意义.方法 分别采用荧光实时定量PCR法和免疫组化法检测60例大肠癌组织及远癌切端大肠组织中的VEGF-C、STAT3 mRNA及其蛋白.结果 大肠癌组织组织中VEGF-C、STAT3 mRNA表达量分别为0.81±0.33、1.33 +0.48,VEGF-C、STAT3蛋白阳性表达者分别为39例、42例;远癌切端大肠组织中分别为0.62±0.28、1.04 +0.30、15例、4例.两者比较,P均<0.05.大肠癌组织中,VEGF-C、STAT3 mRNA及其蛋白表达呈正相关(r分别为0.982、0.892,P均<0.01).VEGF-C、STAT3 mRNA及其蛋白表达水平与大肠癌肿瘤分化程度、浸润深度、淋巴结转移及Dukes分期有关(P均<0.05).结论 大肠癌组织中STAT3、VEGF-C mRNA 及其蛋白高表达.二者在大肠癌的淋巴结转移中发挥重要作用.%Objective To investigate the mRNA transcription and protein expression of vascular endothelial growth factor C( VEGF-C) and signal transducers and activators of transcription 3(STAT3) in colorectal cancer, and study the clinical significance. Methods The levels of VEGF-C, STAT3 mRNA transcription and protein expression of carcinoma and surgical margin tissues gotten from 60 cases with CRC were determined by real-time PCR and immunohistochemical strain separately. Results The VEGF-C, STAT3 mRNA transcription values of tumor tissues were 0. 81 ±0. 33, 1. 33 ± 0. 48, and VEGF-C, STAT3 protein positive expression were detected in 39 and 42 cases; the values in surgical margin tissues were 0.62 ± 0.28, 1.04 ± 0.30 and 15、4 cases separately, P < 0.05. In colorectal cancer tissue , the correlation of mRNA transcription and protein expression were positive (values of r were 0.982、0. 892 separately, all P < 0.01). The levels of VEGF-C、STAT3 mRNA transcription and protein expression were related with differentiation of colorectal cancer, the depth of tumor, lymphatic node metastasis as well as Dukes stage (all P<0.05). Conclusions The STAT3, VEGF-C mRNA transcription and protein express highly in colorectal cancer tissues. STAT3, VEGF-C play an important role in lymph metastasis of colorectal cancer.
