铁载体产生菌Paenibacillus illinoisensis YZ29在花生根际定殖能力研究

         

摘要

利用绿色荧光蛋白(GFP)标记靶微生物是目前研究微生物和宿主相互作用的重要手段.在本研究中,研究者用电击转化的方法将穿梭载体pGFP4412导入铁载体产生菌伊利诺伊类芽孢杆菌(P.illinoisensis)YZ29中,并得到成功表达GFP的YZ29-gfp菌株.利用双抗平板筛选并结合荧光显微镜观察的方法检测了铁载体产生菌YZ29在花生根部及土壤中定殖情况.结果表明:标记菌株在激发光波长为488 nm的蓝光下可观察到绿色荧光;盆栽情况下YZ29-gfp可以在花生根际土和非根际土中定殖;实验室培养条件下其在花生根表定殖,在根内部没有定殖.说明,YZ29能在花生根际有效定殖,为其促生和生防作用奠定了基础.%Marking target microorganisms with green fluorescent protein (GFP) is an important means of studying the interaction between microorganisms and hosts.In this study,the shuttle vector pGFP4412 was introduced into the siderophore-producing bacterium Paenibacillus illinoisensis YZ29 by electroporation,and the YZ29-gfp strain successfully expressing GFP was obtained.The colonization of YZ29 on the peanut root surface and in the soil was studied by double-resistant plate screening combining with fluorescence microscope observation.The results showed that the green fluorescence could be observed under the blue light with excitation wavelength of 488 nm.The YZ29-gfp could colonize in the peanut rhizosphere and non-rhizo-sphere soils under pot culture.Under the condition of laboratory culture,it could colonize on the peanut root surface and could not colonize in the root.This showed that YZ29 could effectively colonize in the peanut rhizosphere,which layed a foundation for growth-promoting and biocontrol effects of peanut.
