一株耐冷产甲烷菌的分离及其生长特性初步研究

         

摘要

以实验室某长期稳定运行的UASB反应器的污泥为样品来源,通过降温筛选、分离纯化,得到一株在15℃条件下生长良好的产甲烷菌株US-3,在荧光显微镜下观察其菌落发出蓝色荧光,高倍扫描电镜观察菌体为较规则球状.经正交试验对其生长特性进行研究,试验结果表明,影响该产甲烷菌株生长能力的因素按主次依次为底物种类、温度、pH值、Na+浓度,各因素最佳水平为底物甲酸钠,温度25℃,pH值6.5,Na+浓度0.6mol· L-1.%A strain of methanogen which grew well at a low temperature of 15 ℃ was obtained from anaerobic sludge in a UASB reactor by selecting, separating and purifying. The colony emitted blue fluorescent when observed by fluorescence microscope. Their colonies were regular globe when observed by high-powered scanning electron microscope. The methanogen's growth charac teristics were studied by an orthogonal experiment. The results showed that the influence of envi ronmental factors on the methanogen's growth was sorts of substrate >temperature>pH value con centration of Na+, and the preferable levels of these factors were sodium formate used as substrate, temperature of 5 ℃, pH 6.5, and 6.5 mol N+·L-1 accordingly.
