CHEMICAL SYNTHESIS AND CLONING OF A GENE FOR YEAST tRNAGly

         

摘要

<正> A 169 bp gene for the yeast tRNAGly has been chemically synthesized.The gene contains a tran-scripting promoter and a terminator at the upstream and the downstream,respectively,of the coding se-quence for yeast tRNAGly.Twe restriction enzyme sites,EcoRI and PstI sites,have been put at bothends of the gene.The process of synthesis involved the preparation of 13 oligodeoxynucleotides andenzymatic ligation.After ligating the synthetic gene to the phage vector M 13mp18 and transformation of itinto JM103 strain,a clone containing the anticipated gene sequence was obtained by hybridization.Theaccuracy of its sequence was confirmed by DNA sequencing.
