六肽对犬血小板聚集功能的影响

         

摘要

Objective This study was aimed to investigate the aggregation effect of hexapeptide on dog platelets .Methods Arteria cruralis blood of dog was sampled and anticoagulated with sodium citrate .The platelet aggregation rates induced by different in‐ducers were determined with the method of turbidimetric test .Results The inhibition rates of dog platelet aggregation of 1 × 10-6 mol・L -1 hexapeptide induced by ADP ,arachidonic acid and thrombin ,were 88 .47% ± 1 .15% ,97 .68% ± 0 .72% and 32 .02% ± 3 .78% ,respectively .The aggregation rates of dog platelet aggregation of 1 × 10-8 to 1 × 10-5 mol・L -1 hexapeptide induced by ADP were significantly lower than that of control group(P<0 .01 or P<0 .05) ,and the corresponding aggregation rates of each dosage groups had significant difference(P< 0 .01 or P< 0 .05) .The anti‐platelet aggregation IC50 of hexapeptide induced by arachidonic acid ,ADP and thrombin ,were 1 .29 × 10-9 ,7 .24 × 10-8 and 2 .88 × 10-6 mol・L -1 ,respectively .For eptifibatide , the anti‐platelet aggregation IC50 were 6 .76 × 10-10 ,5 .74 × 10 -8 and 1 .62 × 10 -6 mol・L -1 ,respectively .Conclusion Hexapep‐tide can restrain dog platelet aggregation in vitro ,and the effect is similar to eptifibatide .%目的:研究六肽对犬血小板聚集功能的影响。方法采集犬股动脉血,以枸橼酸钠抗凝,用比浊法测犬血小板在不同诱导剂诱导时的聚集率。结果1×10-6 mol・L -1六肽,抑制由二磷酸腺苷、花生四烯酸和凝血酶诱导的犬血小板聚集的抑制率分别为88.47%±1.15%,97.68%±0.72%和32.02%±3.78%;六肽1×10-8~1×10-5 mol・L -1组对由ADP诱导的血小板聚集率显著低于对照组( P<0.01或 P<0.05),且各剂量对之间相应的血小板聚集率有显著差异( P<0.01或 P<0.05)。六肽抑制犬由花生四烯酸、二磷酸腺苷和凝血酶诱导的血小板聚集的IC50分别为1.29×10-9,7.24×10-8和2.88×10-6 mol・L -1;对于依替巴肽,同等条件下测得其IC50分别为6.76×10-10,5.74×10-8和1.62×10-6 mol・L -1。结论六肽在体外可以抑制犬血小板聚集,其作用效果类似于依替巴肽。
