泄浊化瘀降脂灵对高血脂症大鼠肝脏ICAM-1、MCP-1表达的影响

         

摘要

目的 通过观察泄浊化瘀降脂灵对高血脂症大鼠的血脂以及肝脏黏附分子-1(ICAM-1)、单核细胞趋化因子-1(MCP-1)表达的影响,探讨泄浊化瘀降脂灵对高脂血症大鼠肝脏的保护机制.方法 将大鼠随机分成空白组、模型组、辛伐他汀组以及浊化瘀降脂灵高、低剂量组,采用高脂饲料复制高血脂症大鼠模型.应用酶法检测血中TC、TG、HDL-C、LDL-C的含量,光镜观察肝脏形态学的改变,免疫组织化学法观察肝组织细胞间ICAM-1、MCP-1的蛋白表达.结果 ①与空白组比较,模型组造模后TC、TG、LDL-C水平显著升高(P<0.05或0.01),HDL-C水平显著降低(P<0.01),提示各组大鼠造模成功.②与〗模型组比较,泄浊化瘀降脂灵高剂量组、辛伐他汀组TC、TG及LDL-C水平显著降低(P均<0.01),HDL-C水平显著升高(P<0.01).泄浊化瘀降脂灵高、低剂量组比较亦有显著性差异(P均<0.05).③与模型组比较泄浊化瘀降脂灵高剂量组及辛伐他汀组肝脏ICAM-1、MCP-1的OD值均显著降低(P均<0.01).泄浊化瘀降脂灵高剂量组与辛伐他汀组比较无显著性差异(P均>0.05).结论 泄浊化瘀降脂灵高剂量可下调MCP-1及ICAM-1高表达,减少、减轻肝细胞脂肪变性和炎症反应,保护高血脂症大鼠的肝脏.%Objective It is to observe the influence of Xiezhuo Huayu Jiangzhiling ( XZHYJZL ) on the lipid metabolism and the expression of ICAM - 1 and MCP - 1 in liver of rat with hyperlipidemia, so as to investigate the protection mechanism of XZHYJZL on liver of rat with hyperlipidemia. Methods 60 healthy male rats were divided into 5 groups ( 12 rats every group ) : blank group, model group, simvastatin group, XZHYJZL high group and XZHYJZL low group. The contents of serum TC , TG ,LDL - c and HDL - c were measured by enzyme method; the change of the liver morphology; and the expressions of protein in hepatic were observed by optical electron-microscopy, the expressions of ICAM - 1 and MCP - 1 in hepatic were obwere increased remarkahly ( P < 0. 01 ) , and the level of HDL - C was decreased ( P < 0. 01 ) , which indicated it was successful to form the animal model.②Compared with the model group, the levels of TC , TG and LDL - C in XZHYJZL high group,simvastatin group were decreased remarkably ( P <0. 01 ),and the level of HDL - c was increased remarkably ( P <0. 01 ).But there was significant difference between XZHYJZL high and low does group ( P < 0. 05 ). ③Compared with the model group, the OD value of ICAM - 1 and MCP - 1 in XZHYJZL high group and simvastatin group were decreased remarkably ( P <0. 01 ). But there was no significant difference between XZHYJZL high does group and simvastatin group. Conclusion XZHYJZL high dose can decrease the high expression of ICAM - 1 and MCP - 1, alleviate the fattiness cell denaturalization and inflammation of liver, and protect the function of liver.
