Detection of miR-21, -122 and -222 contents in serum and tissues of patients with renal cell carcinoma and exploration of their related target genes

         

摘要

Objective:To study the miR-21, -122 and -222 contents in serum and tissues of patients with renal carcinoma and explore their potential target genes.Methods: A total of 54 cases of patients who were diagnosed with renal carcinoma and received surgical resection in Shenzhen Second People''s Hospital between May 2014 and December 2016 and 70 healthy volunteers who received physical examination during the same period were selected as the renal carcinoma group and control group respectively. Then serum samples, renal carcinoma tissues and paracancerous tissues were collected. The expression levels of miR-21, -122 and -222 in serum, renal carcinoma tissues, paracancerous tissues as well as the expression levels of tumor suppressor genes and invasion suppressor genes in renal carcinoma tissues and paracancerous tissues were determined.Results: Serum miR-21, -122 and -222 expression of renal carcinoma group were significantly higher than those of control group;miR-21, -122 and -222 expression in renal carcinoma tissues were significantly higher than those in paracancerous tissues while PDCD4, BTG1, NPRL2, Bax, TIMP1, TIMP2 and E-cadherin expression were lower than those in paracancerous tissues. miR-21, -122 and -222 expression were negatively correlated with PDCD4, BTG1, NPRL2, Bax, TIMP1, TIMP2 and E-cadherin expression. Conclusion:High expression of miR-21, -122 and -222 in serum and tissues of patients with renal carcinoma can inhibit the expression of a variety of tumor suppressor genes and invasion suppressor genes so as to promote the proliferation and invasion of renal carcinoma cells.
