A-FABP与PTEN的相关性以及两者与肥胖、胰岛素抵抗的相关性研究进展

     

摘要

脂肪细胞型脂肪酸结合蛋白(A-FABP)是脂肪酸结合蛋白家族中的一员,主要是在脂肪细胞和巨噬细胞中表达,A-FABP 基因缺陷可改善胰岛素抵抗,并抑制动脉粥样硬化的发生和发展.A-FABP已经成为治疗糖尿病和动脉粥样硬化的重要潜在靶标.第10号染色体缺失的磷酸酶张力蛋白同源物(PTEN)是第一个被发现的具有磷酸酶活性的抑癌基因,它抑制PI3K-Akt信号转导通路,调控胰岛素分泌信号通路.研究显示PTEN与A-FABP在分化的前脂肪细胞3T3-L1中有相互作用.A-FABP是否参与糖代谢途径,是否与PTEN共同调节胰岛素信号转导途径有待进一步研究.%Adipocyte fatty acid binding protein( A-FABP ) belongs to a family of fatty acid binding proteins, which is mainly expressed in adipocyte and macrophage. It has been shown that loss of A-FABP may improve insulin resistance and inhibit the occurrence and development of atherosclerosis. Thus, A-FABP has been considered as a potential therapeutic target in treatment of diabetes and atherosclerosis. PTEN is the first discovered anti-oncogene with phosphatase activities. By the inhibition of PI3 K-AKT signaling pathway, PTFJN regulates the signaling pathways of secretion of insulin. A recent finding provides an evidence of PTFJN and A-FABP interaction in differentiated 3T3-L1 cells. However, the role of A-FABP in glycometabolism and the regulation of insulin signaling pathway with PTEN need further studies.
