血流动力学变化对大鼠颈动静脉移植血管重构影响的研究

     

摘要

目的:血流动力学差异对大鼠动静脉移植后血管重构的影响。方法取40只SD大鼠,随机分为实验组和对照组,每组各20只。实验组取大鼠左侧颈总动脉1 cm移植到大鼠右侧颈外静脉;对照组取大鼠左侧颈总动脉1 cm切除后原位移植。分别于术后第1、2、4周进行大体观察、血管超声检查、血管内膜厚度观察、细胞增生情况观察、细胞凋亡情况观察。结果两组大体观察血流均通畅。超声检查显示实验组血流信号较弱。实验组各观察时间点血管内膜厚度均低于对照组(P<0.05),术后第1、2周两组增殖细胞核抗原(PCNA)指数比较差异有统计学意义(P<0.05),两组各观察时间点凋亡指数比较差异有统计学意义(P<0.05)。结论大鼠动脉血管移植后,血流动力差异导致血管内膜呈现萎缩和增生不同结果,细胞的凋亡和增生发挥了重要作用。动脉血管移植到静脉后,血管重构符合静脉血流动力学特点。%Objective To explore the effects of hemodynamic differences on the vascular reconstruction after cer-vical arteriovenous transplantation in rats. Methods A total of 40 SD rats were randomly divided into experimental group (n=20) and control group (n=20). In experimental group, 1cm of left common carotid artery was transplanted into the right external jugular vein; in control group, 1cm of left common carotid artery was resected and transplanted in situ. General observation, vascular ultrasound, observations of vascular intimal thickness, cellular hyperplasia and cellular ap-optosis were performed in postoperative 1 st , 2 nd and 4 th weeks. Results The blood flow were smooth during general ob-servation in the both groups, and ultrasonography showed that the signal of blood flow in experimental group was weaker than that in control group. The values of intimal thickness on all time points in experimental group were significantly lower than those in control group (P<0. 05), and the differences in proliferating cell nuclear antigen (PCNA) indexes in post-operative 1st and 2nd weeks were statistically significant (P<0. 05), and the differences in apoptotic indexes in all time points were also statistically significant ( P<0. 05 ) . Conclusion After arterial transplantation, hemodynamic differ-ences lead to intimal atrophy or hyperplasia, in which cellular apoptosis and hyperplasia play an important role. After the artery being transplanted into venous, vascular remodeling in venous is in accord with hemodynamic characteristics.
