MALDI-TOF-MS技术快速鉴定临床酵母样真菌的应用评价

         

摘要

目的:评价基质辅助激光解析/电离飞行时间质谱(MALDI‐TOF‐MS)在快速鉴定临床分离的酵母样真菌中的应用价值。方法收集临床分离的酵母样真菌135株,分别用 VITEK MS 、API20C 和 CHROMargar 显色培养同时进行鉴定与比较,χ2检验分析 VITEK MS 和 API 及显色培养基鉴定结果的准确率,以真菌内转录间隔区(ITS)基因测序作为鉴定结果的金标准,同时比较各种方法在临床使用中的优缺点。结果 VITEK MS 、API20C和显色培养基鉴定的准确率分别为98.5%(133/135)、78.5%(106/135)、59.3%(80/135),经χ2检验分析,差异有统计学意义(χ2=51.996,P<0.05)。结论 MALDI‐TOF‐MS 可将酵母样真菌快速、准确地鉴定到种。%Objective To evaluate the application value of matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry(MALDI‐TOF‐MS) for rapid identification of clinically isolated yeast‐like fungi .Methods 135 strains of clinically isolated yeast‐like fungi were collected and identified by VITEK MS ,API20C system andCHROMagar Candida at the same time ,then the detection results were compared ,the accuracy of detected resultswere analyzed by the χ2 test ,the identification of the ITS gene sequence of these isolates were taken as the goldstandard .At the same time the advantages and disadvantages in the clinical application of these methods were compared.Results The identification accuracy rates of VITEK MS ,API20C and CHROMagar Candida were 98 .5%(133/135) ,78 .5% (106/135 ) and 59 .3% (80 /135 ) respectively ,the differences were statistically significant (χ2 =51 .996 ,P< 0 .05) .Conclusion MALDI‐TOF‐MS could rapidly and accurately identify yeast‐like fungi .
