红腺忍冬叶抗氧化作用的实验研究

         

摘要

[目的]研究红腺忍冬叶的抗氧化作用,为红腺忍冬叶的开发利用奠定一定的实验依据.[方法]时红腺忍冬叶不同溶剂提取物进行体内外抗氧化实验研究:(1)以Vc、二丁基羟基甲苯(butylated hydroxytoluene,BHT)为对照品,通过1,1-二苯基-2-三硝基苯肼(1,-diphenyl-2-picrylhy drazyl,DPPH)法和铁离子还原法考察样品的体外抗氧化活性.(2)以Vc为阳性时照,由D-半乳糖至衰的小鼠为模型,通过测定血清、肝脏组织中超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘过氧化酶(glutathione peroxidase,GSH-PX)活性和丙二醛(malondialdehyde,MDA)含量,考察样品的体内抗氧化活性.[结果](1)在0.5~2mg·mL-1范围内,水提液、50%乙醇提取液和95%乙醇提取液对DPPH自由基的清除效果与Vc无显著性差异或优于Vc,多与BHT尚有差距,三种提取液之间无显著性差异(P>0.05);在0.1~5mg·mL-1范围内,三种提取液对Fe3+的还原能力随着浓度上升显著增强,但与Vc、BHT尚有差距,三种提取液之间无显著性差异(P>0.05).(2)综合SOD结果:水提液高剂量组作用优于其他各药物组,且其血清SOD活力与Vc组无显著性差异(P>0.05),肝脏中SOD活力只有该组与模型组比较有显著性效果.综合GSH-PX活性结果:除50%醇提高低剂量组外,其余各组均具有较好活性;其中水提液高剂量组在血清中的GSH-PX活性显著高于Vc组.综合MDA结果:水提液高低剂量组和50%、95%醇提高剂量组均具有较好的效果,而且这些组在血清和肝脏中的MDA含量与Vc组相比,降低无显著性差异(P>0.05).[结论]红腺忍冬叶的不同溶剂提取物都具有一定的体内外抗氧化效果,特别是对提高GSH-PX活性和对MDA的清除效果较强.其中水提液抗氧化效果优于醇提物,值得深入研究.%[Objective] To study the antioxidant effect on leaves of Lonicera Hypoglauca Miq.and provide an experimental basis for the development and utilization of leaves of Lonicera Hypoglauca Miq.. [Methods] Antioxidant activities of leaves of Lonicera Hypoglauca Miq. were studied by the experiments in vivo and in vitro of different extracts. ① Compared with Vc and BHT, the vitro antioxidant activities were studied by 0PPH radical scavenging and the reducing capacity of Fe3+. ② Senile mouse models induced by D-galactose were used. Compared with Vc, the vivo antioxidant activities of extracts were studied by measuring SOD, GSH-PX activity and MDA content of serum and liver tissue. [Results] ①In 0.5~5mg·mL-1, the scavenging effect of water extract, 50% ethanol extract and 95% ethanol extract on DPPH free radicals had no significant difference or the more excellent performance with that of Vc but mostly a gap with that of BHT. Three extracts showed no significant difference from one another. In 0.1~5mg·mL-1 the reducing capacity of three extracts on Fe3+ significantly enhanced with the increasing of concentration, but had a certain gap with that of BHT and Vc. Three extracts showed no significant dif-ference from one another. ②The comprehensive results of SOD: the high dose group of water extract had a better effect than the other drug groups, Its SOD activity in the serum had no significant difference with that of VC group. For the SOD activity in the liver, only this group showed a significant effect com-pared with the model group. Comprehensive GSH-PX activity results: except high and low dose groups of 50% ethanol extract, the other groups all had good activity. Among which in the high dose group of water extract, GSH-PX activity in the serum was significantly higher than that of VC group. Inte-graced MDA results: high and low dose groups of water extract, high dose groups of 50% ethanol extract and 95% ethanol extract had good effects. And in these groups, the MDA content in the serum and liver had no significant difference or significant reduction compared with that of VC group. [Conclusion] The different extracts of leaves of Lonicera Hypoglauca Miq.had antioxidant activity both in vitro and in vivo, especially obviously increasing GSH-PX ac-tivity and remarkably removing effect of MDA. The water extract had the more excellent performance compared with ethanol extract Antioxidant effect of water extract on leaves of Lonicera Hypoglauca Miq. is worthy of further research.
