黑果小檗提取物对高脂血症大鼠血脂、血液流变学和脂质过氧化水平的影响

         

摘要

目的 研究黑果小檗提取物对高脂血症大鼠血脂、血液流变学和脂质过氧化水平的影响.方法 将50只SD大鼠随机分成模型组,辛伐他汀组(5mg/kg),黑果小檗提取物低、中、高剂量组(100、200,400mg/kg),每天灌胃给予高脂乳剂(10 mL/kg),连续灌胃20 d;另取10只SD大鼠作为正常对照组,灌胃给予蒸馏水(10mL/kg),连续灌胃20 d.各组大鼠给药6 w后腹主动脉取血,全血采用全自动血流变仪测定全血粘度、血浆粘度、红细胞压积、聚集指数、变形指数、刚性指数,分离血清采用全自动生化仪测定胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C),并采用全自动酶标仪测定丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA).结果 与模型组比较,黑果小檗提取物低、中、高剂量组大鼠血清TC、TG、LDL-C、ALT、AST、全血粘度、血浆粘度、红细胞压积、MDA降低,HDL-C、红细胞变形能力、SOD、GSH-Px升高,差异有统计学意义(P<0.05),其中黑果小檗提取物高剂量组作用最为明显:TC(2.87±0.36) mmol/L、TG(0.67±0.06) mmol/L、LDL-C(0.93±0.19) mmol/L、ALT(42.28±6.36)U/L、AST(89.25±9.31)U/L、全血粘度-低切(8.22±1.96) mPa/s、全血粘度-中切(3.88±0.86 mPa/s)、全血粘度-高切(3.45±0.65) mPa/s、血浆粘度(1.19±0.12)mPa/s、红细胞压积(52.49±7.56)%、MDA(5.19±0.46) mmol/L、HDL-C(0.95±0.13)mmol/L、SOD(183.64±22.51)U/L、GSH-Px(42.80±6.36) pmol /L.结论 黑果小檗提取物对高脂血症大鼠具有降血脂作用,可改善血液流变学并减轻脂代谢紊乱的过氧化损伤.%Objective To study the activity of Beberis heteropoda extracts on blood lipid,hemorheology and antioxidation of hyperlipidemia rats.Methods 50 SD rats were divided into 5 groups as model group,simvastatingroup (5 mg/kg),low (100 mg/kg),medium (200 mg/kg) and high (400 mg/kg) dose of Beberis heteropoda groups,given high fat emulsion (10 mL/kg) by intragastric administration for 20 d,10 SD rats were given distilled water for normal control group.After 6 weeks of treatment,whole blood viscosity,plasma viscosity,hematokrit,indexes of erythrocyte aggregation,deformation and rigidity by automatic blood rheometer.The levels of TC,TG,HDL-C and LDL-C in serum were tested by automatic biochemical analyzer.The levels of ALT,AST,SOD,GSH-Px and MDA were tested by microplate absorbance reader.Results Compared with model group,the levels of TC,TG,LDL-C,ALT,AST,MDA,blood viscosity,hematokrit,erythrocyte aggregation and rigidity index were decreased,the levels of HDL-C,SOD,GSH-Px and erythrocyte deformation index were increased (P <0.05).High (400 mg/kg) dose ofBeberis heteropoda group showed the most significant effect:TC(2.87±0.36)mmol/L,TG(0.67±0.06) mmol/L,LDL-C(0.93±0.19)mmol/L,ALT(42.28±6.36)U/L,AST(89.25±9.31)U/L,blood viscosity-low shear(8.22±1.96)mPa/s,blood viscosity-middle shear(3.88±0.86)mPa/s,blood viscosityhigh shear(3.45±0.65)mPa/s,plasma viscosity(1.19±0.12)mPa/s,hematokrit(52.49±7.56)%,MDA (5.19±0.46)mmol/L,HDL-C(0.95±0.13)mmol/L,SOD(183.64±22.51)U/L,GSH-Px(42.80±6.36) pmol/L.Conclusion Beberis heteropoda extracts have activities of regulating blood-lipid,improving hemorheology and inhibiting peroxidation damage in hyperlipidemia rats.
