肺吸入胰岛素气雾剂对实验性糖尿病大鼠的治疗作用

         

摘要

Objective To observe the effect of reducing glycemia on rats suffering diabetes melli tus(DM). Methods We induced rats suffering DM by subcutaneous injecting tetraoxypyrimidine.Natrium glycocholate and natrium glycerate were chosen as respiratory tract musoca inhalant en hancer, and were made into inhaled insulin aerosol each other. The aerosol including natrium glyco cholate and natrium glycerate were administered to rats suffering DM after catheterization of bron chus. The level of glycemia was tested. Results We confirmed that rats suffering DM was a lowrnsecretary insulin DM model. The aerosol that natrium glycocholate as inhalant enhancer acted in 15min and peaked in 30min after administration. The aerosol that natrium glycerate as inhalant en hancer acted in 15min and peaked in 2h after administration. The group subjected to subcutaneous injection insulin peaked in 1h after administration. The aerosol mentioned above were administrated to rats with DM in different dose of 5IU/kg, 10IU/kg, 15IU/kg. We observed that there was no significant enhancement of glycemia decreasing percent. Conclusion The pulmonary inhaled insulin aerosol which we made can obviously reduce glycemia.%目的观察肺吸入胰岛素气雾剂对糖尿病大鼠血糖的影响。方法用四氧嘧啶静脉注射诱导糖尿病大鼠模型,以甘氨胆酸钠及甘珀酸钠为吸收促进剂,分别制成肺吸入胰岛素气雾剂,糖尿病大鼠气管插管后用药,测定血糖。结果糖尿病大鼠模型为低胰岛素分泌型。甘氨胆酸钠作吸收促进剂的气雾剂组在给药后15min起效,30min血糖下降百分数达峰值;甘珀酸钠为吸收促进剂的气雾剂在给药后15min起效,2h血糖下降百分数达峰值;对照的皮下注射胰岛素组在用药后1h血糖下降百分数达峰值。气雾剂用量增加,糖尿病大鼠血糖下降百分数无明显增加。结论肺吸入胰岛素气雾剂有明显的降血糖作用。
