罗格列酮对OLETF鼠皮肤组织及其晚期糖基化产物的影响

             

摘要

Objective: To research the effect and mechanism of rosiglitazone on the skin and AGEs in OLETF rats. Methods: 20 male OLETF rats were given high-calorie diet for 30 weeks to set up the T2DM. At the 30th week, 12 male induced OLETF rats were randomized into two groups: model group and rosiglitazone group 8 LETO rats were used as controls. The shaved skin specimens from the back of rats in three groups were collected. Skin AGEs concentrations were assessed by immunohistochemistry assay. Hematoxylin-eosin dye was used for histological examination. Masson trichrome staining was used for the collagen fiber. Results: The skin thickness in model group was decreased. The features of multilayer epithelium structure disappeared in epidermis. The dermis layer became thinner with tenuous collagen distributing disorderly and part of the collagen fibers in dermis became atrophic, swollen and degenerated. However such changes were not detected in rosiglitazone group. In model group, the expression of AGEs increased significantly compared to rosiglita- zone group and control group. Conclusion: A histochemical changes have already been occurred in diabetic skin before injury , which may be result from AGEs. Rosiglitazone may postpone the course of diabetic skin lesions to reduce the nuisance.%目的:探讨罗格列酮对OLETF鼠皮肤组织及其晚期糖基化产物(AGEs)的影响和机制.方法:OLETF雄性大鼠20只,30周时共有成模的2型糖尿病(T2DM)OLETF大鼠12只,随机分为罗格列酮组和模型组(每组6只),对照组为LETO鼠(8只).各组均于给药12周后同时采集背部皮肤组织,HE染色观察皮肤的组织学变化及测量表皮和真皮的厚度,Masson三色染色观察胶原纤维的变化,免疫组织化学染色观察AGEs的蓄积程度及变化.结果:模型组大鼠表皮组织变薄,表皮细胞欠清晰,部分表皮缺乏复层排列;真皮层亦变薄,胶原纤维纤细、排列紊乱,部分胶原可见肿胀、变性.罗格列酮组大鼠皮肤表皮层较清晰,复层排列接近对照组,真皮胶原丰富,表皮与真皮厚度明显增加,与对照组无明显差异.模型组大鼠皮肤组织AGEs明显蓄积在真皮基质和细胞中,阳性表达明显高于罗格列酮组和对照组.结论:2型糖尿病在皮肤未受损伤时已发生组织学变化,AGEs可能是导致这种变化的机制之一,而罗格列酮可有效减轻这种损害的发生.
