串叶松香草组培快繁技术研究

             

摘要

The influence of different hormones concentration composition on the tissue culture capability of different explants (tender shoot, young leaves and leafstalk ) in Silphium perfoliatum L was analysed. The result was listed as below: the callus formation capability of different explants is different, the tender shoot was better than leafstalk and the leafstalk better than yoong leaves. The optimum culture media were established, callus induction medium was MS ( Murashige & Skoog ) basal medium supplemented with 6-BA 2.0 mg/L and 2,4-D 3.0 mg/L, proliferation medium was MS supplemented with 6-BA 2.5 mg/L and IBA 0.5 mg/L, rooting medium was 1/2 MS supplemented with 6-BA 0.2 mg/L and IBA 2.0 mg/L. When the root was about five centimeters long, the culture flasks were opened for 2 ~ 3 days and transplanted to the pot with peat, perlite and vermiculite, the ratio of bulk material was 1∶ 1∶ 1. One month later, over ninety percent of the transplanted plantlets were alive.%以串叶松香草的嫩芽、新叶和叶柄的切块为外植体,分析在不同上培养阶段不同激素浓度配合使用对其组培能力的影响.结果表明,不同外植体诱导形成愈伤组织的能力大小依次为嫩芽>叶柄>新叶;串叶松香草的最适诱导培养基为MS+6-BA2.0mg/L+2,4-D 3.0mg/L,增殖培养基为MS+6-BA 2.5 mg/L+IBA 0.5 mg/L,生根培养基为1/2 MS+6-BA 0.2 mg/L+IBA 2.0 mg/L;当根系长约5 cm时,开瓶炼苗2~3 d,移栽至装有基质(V(泥炭)∶V(珍珠岩)∶V(蛭石)=1∶1∶1)的花盆内炼苗1个月后即可移入大田,移栽成活率达90%以上.
