多囊卵巢综合征中卵巢初级小卵泡bcl-2、bax的表达

         

摘要

Objective: The aim of the study was to investigate the expression and implication of the apoptosis regulatory protein bcl-2 and bax in different classes of primary follicles of polycystic ovary syndrome(PCOS).rn  Methods: The study groups consisted of 18 PCOS and 13 normal subjects collected from March 1997 to May 1998. Total numbers of follicles collected from their ovaries were 240, which were divided into four groups based on diameter of the follicles. The apoptosis regulatory protein bcl-2 and bax were assayed using immunohistochemical method and the grey value by Germany Konton Ibas Software was used to judge the results quantitatively. Analysis of variance was performed using SPSS Package.rn  Results: The bcl-2 protein expression in different classes of preantral follicles from PCOS groups was not significantly different, however, the bax protein expression in different classes showed to have significant difference. When the diameter of the follicle reached to 60~120 μm, bcl-2 expression was significantly higher in PCOS than in control group (P<0.05).Bax expression was increased markedly as primary follicles grew in PCOS, whereas it was decreased when the diameter of follicle reached >120 μm, and bcl-2/bax grey ratio was apparently decreased as well.rn  Conclusion: The bcl-2 and bax protein expression in different classes of primary follicles were significantly higher in PCOS than that in normal ovary when follicle in diameter was 60~120 μm. The bcl-2 and bax protein expression increased following follicle growth in PCOS and control groups. When the diameter of follicle reached >120 μm, the apoptosis was weakened resulting in accelerating growth of follicle. This study suggests that before primary follicles, the expression of apoptosis regulate protein bcl-2 and bax were normal in PCOS.%目的:探讨凋亡调节蛋白bcl-2及bax在多囊卵巢综合征(PCOS)患者的卵泡选择、发育的作用。方法:18例PCOS患者及13名正常人240个小卵泡按直径分为4个组,采用免疫组织化学方法及Konton Ibas灰度值的定量测定,用SPSS软件统计,定量分析凋亡调节蛋白bcl-2及bax在PCOS各级初级小卵泡中的表达。结果:PCOS组各级窦前卵泡组间bax蛋白表达有显著差异,随着卵泡直径的增加bax表达明显增多,但直径>120 μm的卵泡bax表达减少,bcl-2/bax灰度比值显著变小。bcl-2蛋白表达相对稳定:PCOS组直径在60~120 μm的卵泡bcl-2表达显著高于正常对照组(P<0.05)。结论:正常人及PCOS卵巢组织各级窦前卵泡和窦卵泡均表达bcl-2及bax,随着卵泡的增大,bcl-2的表达增加,当卵泡直径>120μm时,细胞凋亡减弱,卵泡的增长加速。PCOS患者卵巢中各级窦前卵泡和窦卵泡凋亡蛋白bcl-2及bax的表达是正常的。
