青岛百合高效离体快速繁殖体系的建立

         

摘要

以青岛百合及麝香百合的鳞片和茎尖为试验材料,研究了外植体的放置方式、鳞片的不同部位、不同的激素浓度及活性炭对百合再分化的影响,成功建立了青岛百合的高频再生体系.结果表明两种百合的最佳外植体部位为中层鳞片的基部,放置方式为凸面接触培养基,青岛百合的最适再生培养基为MS+6- BA1.5mg/L+ NAA0.2mg/L,麝香百合的最佳再生培养基为MS+6- BA l.0mg/L+ NAA0.lmg/L,两种百合的最高再生率均达到100%.%Tissue culture and rapid propagation on Lilium tsingtauense Cilglilia Ceae and Llilium longiflorum using bud scales of musk were studied. Through inducing adventitious bud, several factors such as different placements of scale, different hormones affecting bud regeneration were discussed. Results showed that the appropriate part of scale for inducing the adventitious bud was the basal part; It was adaptable to set the convexity of scale facing medium when including adventitious buds. MS medium complemented with 6 - BA1. 5mg/L + NAAO. 2mg/L had the best effect on adventitious bud induction in Lilium tsingtauense Cilglilia Ceae, and MS +6 - BA1. Omg/L + NAA 0. Lmg/L was the optimum medium for induction of adventitious bud in Lilium longiflorum, the regeneration rate of both varieties were 100%.
