HLA-G14bp基因多态性及HLA-GmRNA表达水平与原因不明复发性流产的关系

         

摘要

Objective:To investigate the correlation of human leukocyte antigen-G(HLA-G) 14bp gene deletion or insertion and expression of HLA-G mRNA with unexplained recurrent spontaneous abortion (URSA). Methods;39 patients with URSA and 52 women who had one natural childbirth and would have induced Abortion were recuited in this study, the polymorphism of HLA-G 14bp and the expression of HLA-G mRNA in villous tissue in two groups were detected by polymerase chain reaction (PCR) and reverse transcrip-tion-polymerase chain reaction (RT-PCR). Results-. Gene Frequency of HLA-G 14bp heterozygote ( -14bp/+14bp) in URSA group(64.1%) was much higher than that in normal control group(42. 3%) (xz = 6.511, P=0.039). Gene Frequency of HLA-G 14bp homozygous insertion ( +14bp/ + 14bp) in URSA group(7.7%) was lower than that in normal control group(26. 9%) (x2 =5. 425, P = 0. 020). Gene Frequency of HLA-G 14bp homozygous deletion ( -14bp/-14bp) in URSA group (28.2%) was no obvious difference compared with that in normal control group (30. 8%) (x2 =0. 070, P = 0. 791). Expression of HLA-G mRNA ( -3. 018 ±1. 005) in URSA group was lower than that in normal control group( -1. 072 ± 0. 436 )(f=9. 681 ,P=0. 000).Conclusions:HLA-G 14bp gene heterozygote ( -14bp/ + 14bp) increases the occurrence of disease, and the low expression of HLA-G mRNA in prophase villous tissue is strongly correlated with the development of unexplained recurrent spontaneous abortion.%目的:探讨人类白细胞抗原G14帅(HLA-G 14bp)基因缺失或插入多态性与原因不明复发性流产(URSA)的相关性,了解HLA-G mRNA在URSA患者中的表达水平和意义.方法:选择URSA患者39例(URSA组)和有1次以上正常分娩史并行人工流产的妇女52例(正常对照组)为研究对象,采用聚合酶链反应(PCR)检测14bp基因多态性和逆转录-聚合酶链反应(RT-PCR)检测两组人群绒毛组织中HLA-G mRNA的相对表达量,并进行分析.结果:URSA组14bp基因杂合子(-14bp/+ 14bp)的基因频率(64.1%)明显高于正常对照组(42.3%) (X2=6.511,P =0.039);而插入纯合子(+ 146p/ + 14bp)的基因频率(7.7%)低于正常对照组(26.9 %) (X2 = 5.425,P = 0.020);缺失纯合子(-14bp/-14bp)的基因频率(28.2%)与正常对照组(30.8%)比较,差异无统计学意义(X2 =0.070,P=0.791),URSA组HLA-G mRNA表达水平(-3.018±1.005)明显低于正常对照组(-1.072±0.436) (t=9.681,P=0.000).结论:14bp基因为-14bp+ 146p杂合子时增加了发病的机会,早期绒毛中HLA-G mRNA低水平表达与URSA发生密切相关.
