人类精子库精子冷冻最佳方案的探索

             

摘要

Objective To obtain the optimal protocols for sperm freezing by studying different factors influencing the motility of post-thaw. Methods The effects of cryopreservative methods and concentrations of glycerol, egg yolk, and seminal plasma on the motility of post-thaw were compared. Results The rate of post-thaw motility was significantly higher in slow freezing than in fast freezing. The rate of post-thaw motility was significantly higher in the cryoprotectant containing 6% or 7.5% glycerol than in those of 4% or 9%, but there was no significant difference between the two former cryoprotectants.The rate post-thaw motility did not differ significantly among the cryoprotectants containing egg yolk of 5%, 10%, and 15%, but was significantly higher than that of 0%. The rate of post-thaw motility did not differ significantly between seminal plasma of 50% and that of 75% ,whereas was higher than that of 0%. Conclusions Cryopreservative methods and concentrations of glycerol, egg yolk, and seminal plasma have an effect on the post-thaw motility of human sperm. The slow freeze with glycerol final concentration of 6% ~ 7.5%, egg yolk of 5% ~ 15%, and seminal plasma of 50% or 75% is feasible during the process of spermfreezing.%目的:通过对冷冻过程中影响精子冷冻复苏率的不同因素进行研究,以获取最佳的冷冻方案.方法:比较冷冻过程冷冻方法、甘油浓度、卵黄浓度及精浆浓度对精子冷冻复苏率的影响.结果:(1)缓冻法精子冷冻复苏率高于直接冷冻法,差异有显著性;(2)甘油浓度为6%或7.5%的保护剂冷冻复苏率显著高于甘油浓度为4%及9%的保护剂.但前两者间无显著差异;(3)卵黄浓度为5%,10%,15%的保护剂间冷冻复苏率无显著差异,均高于卵黄浓度为0%的保护荆,差异有显著性;(4)50%及75%精浆浓度冷冻复苏率无显著差异,均显著高于0%的精浆浓度.结论:精子冷冻过程中冷冻方法及甘油、卵黄、精浆浓度对精子冷冻复苏率均有影响.采用缓冻法,甘油终浓度6%~7.5%,、卵黄终浓度5%~15%、精浆终浓度50%或75%的精子冷冻方案是可行的.
