高温高湿环境对自发性高血压大鼠IL-2、IL-12及淋巴细胞增殖的影响

         

摘要

Objective To investigate the influence of the high temperature and humidity environment on weight, blood pressure,serum interleukin 2(IL-2),interleukin 12(IL-12)and lymphocyte proliferation rate of rats with spontaneously hypertensive.Methods 16 male WKY rats were selected and randomly divided them into the control group(WKY-CN)and the group of normal blood pressure(WKY-HS). 16 male SHR rats were selected and randomly divided them into control group of high blood pressure(SHR-CN)and the heat stress group(SHR-HS). The rats in the control group were raised at room temperature,while rats in the heat stress group were raised at 32°C,and the humidity was 65%. After 7 days' heat stress,the changes of food consumption,blood pressure and body weight of rats were measured in two groups while the changes of serum IL-2 and IL-12 were monitored by ELISA and the cell proliferation rate was detected through CCK-8 reagent. Results Compared with the control group,the body weight of rats in the heat stress group increased slowly,and the weight of rats with high pressure reduced rather than increasing for 7 days' heat stress. The food consumption of rats in the heat stress group was decreased,and that in the heat stress group was more obvious. With the comparison of the control group,the systolic blood pressure and diastolic blood pressure(P<0.05)of rats in the heat stress group significantly enhanced,while serum IL-2,IL-12 and lymphocyte proliferation rate had no change. IL-12 of SHR rats was lower significantly than that of WKY rats(P<0.05). Although IL-12 of SHR rats was lower than that of WKY rats,the difference did not have statistical significance.Conclusion The environment of high temperature and humidity for seven days can slow down the growth of rats' body weight, increase the blood pressure of rats,and affects the level of IL-2 and IL-12 and the proliferation rate of lymphocytes in rats' serum,which will gradually adjust to the normal state.%目的 探讨高温高湿环境对自发性高血压(SHR)大鼠体重、血压、血清白介素2(IL-2)、白介素12(IL-12)及淋巴细胞增殖的影响.方法 选用雄性正常血压(WKY)大鼠16只,随机分为正常血压对照组(WKY-CN)与正常血压热应激组(WKY-HS);选用雄性SHR大鼠16只,随机分为高血压对照组(SHR-CN)与高血压热应激组(SHR-HS).对照组大鼠在室温下饲养,热应激组大鼠饲养温度为32℃,湿度为65%.经过7d热应激后,测定各组大鼠的食物消耗量、血压及体重变化,用ELISA法测定大鼠血清IL-2及IL-12含量的变化,通过CCK-8试剂检测大鼠淋巴细胞增殖程度.结果 经过7d热应激或室温环境饲养后,WKY-CN、WKY-HS、SHR-CN组大鼠体重均较处理前有所增长(P均<0.01),而SHR-HS组大鼠体重较热应激前下降(P<0.05);WKY-CN组与WKY-HS组比,7d消耗鼠粮差异无统计学意义(P>0.05),而SHR-HS组比SHR-CN组鼠粮消耗减少(P<0.05);WKY-HS及SHR-HS组大鼠热应激后收缩压及舒张压均上升,且SHR-HS组大鼠收缩压与舒张压升高幅度均大于WKY-SH组大鼠(P均<0.05);SHR大鼠血清IL-2、IL-12水平低于WKY大鼠(P均<0.01),但热应激对大鼠血清IL-2、IL-12以及淋巴细胞增殖程度无影响(P>0.05).结论 高血压大鼠热应激后体重下降;并使大鼠的血压升高;但对大鼠血清中IL-2、IL-12水平及淋巴细胞增殖程度无明显影响.
