提高籼稻恢复系花药培养绿苗分化率的研究

         

摘要

As MS medium was control,the effects of M8 and N6 mediums on anther callus induction rate of indica restorer lines were studied,as well as the differentiation rate of indica restorer lines were studied with different concentrations of sorbitol and activated carbon added in medium. The results showed that four indica restorer lines in M8 medium had the highest rate of callus,ranging from 20. 97% to 30. 63%, significantly higher than that in MS medium;while in the N6 medium,the rate of callus was 10. 77%—13. 70%,only Yanhui 559 and Yanhui 888 were significantly higher than that in MS medium. With the increase of concentration of sorbitol,the green plantlets differentiation rate of four indica restorer lines in the M8 differentiation medium increased, reached the highest value with 30 g/L sorbitol, ranging from 30. 57% to 36. 77%, followed by 20 g/L sorbitol, both significantly higher than the treatment without sorbitol. With the increase of concentration of activated carbon,the green plantlets differentiation rate of four indica restorer lines in M8 differentiation medium increased first and then decreased, reached the highest value with 1 g/L,ranging from 21. 38% to 27. 82%,followed by 2 g/L,both significantly higher than the treatment without activated carbon.%以MS培养基为对照,对比分析M8和N6两种培养基对籼稻恢复系花药培养出愈率的影响,并添加不同质量浓度的山梨醇、活性炭,研究其对愈伤组织绿苗分化率的影响.结果表明,4个籼稻恢复系在M8培养基中均具有最高的出愈率,介于20.97%~30.63%,极显著高于MS培养基;而在N6培养基上,出愈率介于10.77%~13.70%,仅盐恢559和盐恢888显著高于MS培养基.随着山梨醇质量浓度升高,4个籼稻恢复系在M8分化培养基上的花药愈伤组织绿苗分化率增加,以30 g/L山梨醇处理最高,介于30.57%~36.77%,其次为20 g/L山梨醇处理,两者均极显著高于不添加山梨醇处理.随着活性炭质量浓度升高,4个籼稻恢复系在M 8分化培养基上的花药愈伤组织绿苗分化率先增加后降低,总体均以1 g/L处理最高,介于21.38%~27.82%,2 g/L处理次之,两者均极显著高于不添加活性炭处理.
