胃癌患者幽门螺杆菌感染情况与肿瘤恶性程度的相关性研究

[ABSTRACT]Objective:To study the correlation between helicobacter pylori infection and tumor malignancy in patients with gastric cancer.Methods:A total of 1 1 9 patients with gastric cancer treated in our hospital between May 2013 and Decem-ber 201 5 were collected and divided into HP-positive group (n =89)and HP-negative group (n =30)according to the combina-tion of helicobacter pylori infection or not.RIA method was used to detect the serum angiogenesis index levels;fluorescence quantitative PCR (RT-PCR)was used to detect the mRNA expression of autophagy genes and proliferation genes in tumor tis-sues.Results:Serum angiogenesis indexes bFGF,VEGF and MMP-9 levels of HP-positive group were significantly higher than those of HP-negative groups;autophagy genes Beclin1,BNIP3,pULK and PI3KC3 mRNA expression in tumor tissues of HP-positive group were significantly lower than those of HP-negative group,pro-apoptotic genes Bax and p53 mRNA expression were significantly lower than those of HP-negative group,and anti-apoptotic genes PIK3CD,CIP2A and I2PP2A mRNA ex-pression were significantly higher than those of HP-negative group.Conclusions:Helicobacter pylori infection can promote an-giogenesis and cell proliferation in lesions of patients with gastric cancer.%目的：：探讨胃癌患者幽门螺杆菌感染情况与肿瘤恶性程度的相关性。方法：收集2013年5月~2015年12月间本院收治的胃癌患者119例，根据是否伴幽门螺杆菌感染分为 Hp 阳性组89例、Hp 阴性组30例。采用放射免疫法检测血清中血管新生指标含量；采用荧光定量 PCR(RT-PCR)检测肿瘤组织自噬基因、增殖基因 mRNA 表达量。结果：Hp 阳性组患者的血清血管新生指标碱性成纤维细胞因子(bFGF)、血管内皮生长因子(VEGF)、基质金属蛋白酶(MMP-9)含量均显著高于 Hp 阴性组患者；Hp 阳性组患者肿瘤组织中自噬基因 Beclin1、BNIP3、pULK、PI3KC3 mRNA 表达量均显著低于 Hp 阴性组患者，促凋亡基因 Bax、p53 mRNA 表达量显著低于 Hp 阴性组患者，抗凋亡基因 PIK3CD、CIP2A、I2PP2A mRNA 表达量显著高于 Hp 阴性组患者。结论：幽门螺杆菌感染能够促进胃癌患者病灶内血管新生以及细胞增殖。
