外周血及胎盘中MyD88表达量与GDM患者胎盘中炎症反应及胰岛素信号转导的相关性

         

摘要

Objective:To study the correlation of MyD88 expression in peripheral blood and placenta with the inflamma-tory response and insulin signal transduction in the placenta of patients with gestational diabetes mellitus(GDM).Methods:The patients with GDM and healthy pregnant women who accepted antenatal care and gave birth in Guangyuan First People's Hospital between March 2015 and June 2017 were selected as the research subjects and enrolled in the GDM group and control group for the study respectively;the peripheral blood was collected before delivery to determine the MyD88 mRNA expression, and the placental tissue was collected after delivery to determine the mRNA expression of MyD 88,inflammatory response mol-ecules and insulin signal transduction molecules.Results:MyD88 mRNA expression levels in the peripheral blood and placenta of GDM group were significantly higher than those of control group,and the MyD88 mRNA expression in the peripheral blood was positively correlated with the MyD88 mRNA expression in the placenta;IL-1β,IL-6,RBP4,Chemerin,Resistin and PTP1B mRNA expression levels in the placenta of GDM group were significantly higher than those of control group whereas IRS1,ISR2,p-PI3K and GLUT4 protein expression levels were significantly lower than those of control group;IL-1β,IL-6, RBP4,Chemerin,Resistin and PTP1B mRNA expression levels in the placenta of GDM group of patients with high MyD88 ex-pression were significantly higher than those of patients with low MyD88 expression whereas IRS1,ISR2,p-PI3K and GLUT4 protein expression levels were significantly lower than those of patients with low MyD 88 expression.Conclusion:The expres-sion of MyD88 in peripheral blood and placenta increase in patients with GDM and the change of MyD88 expression in peripher-al blood could reflect the abnormality of inflammatory response and insulin signal transduction in the placenta.%目的:研究外周血及胎盘中M yD88表达量与妊娠期糖尿病(GDM)患者胎盘中炎症反应及胰岛素信号转导的相关性.方法:选择在本院产检及分娩的GDM 患者及健康孕妇作为研究对象,分别作为研究的GDM 组和对照组;分娩前采集外周血并测定MyD88的mRNA表达量,分娩后采集胎盘组织并测定MyD88、炎症反应分子、胰岛素信号转导分子的mRNA表达量.结果:GDM 患者外周血及胎盘中MyD88的mRNA表达量均显著高于对照组且外周血中MyD88的mRNA表达量与胎盘中MyD88的mRNA表达量呈正相关;GDM 组胎盘中IL-1β、IL-6、RBP4、Chemerin、Resistin、PTP1B的mRNA 表达量均显著高于对照组,IRS1、ISR2、p-PI3K、GLUT4的蛋白表达量显著低于对照组;GDM 组中MyD88高表达量的患者胎盘中IL-1β、IL-6、RBP4、Chemerin、Resistin、PTP1B的mRNA 表达量均显著高于MyD88低表达量的患者,IRS1、ISR2、p-PI3K、GLUT4的蛋白表达量显著低于MyD88低表达量的患者.结论:GDM 患者外周血及胎盘中M yD88的表达升高且外周血中M yD88表达量的变化能够反应胎盘中炎症反应及胰岛素信号转导的异常.
