广西眼镜蛇毒中Natrin蛋白的分离及纯化

     

摘要

Objective: To isolate and purify Natrin from Guangxi cobra venom. Methods: The Natrin from Guangxi cobra venom was sequentially purified by Sephasex G75gel, CM-sepharose CL-6B ion exchange chromatography and Mono S column chromatography on FPLC system. Results: The Natrin purified from Guangxi cobra venom tested by SDS-PAGE and proved its molecular weight was 24. 937 ku and it's N-ter-minal acid sequence was NVDFNSESTRRKKKQKEIVD and C-terminal acid sequence was NIKK. Conclusion: Natrin can be obtained by gel filtration and ionexchange.%目的:从广西眼镜蛇毒中分离出Natrin蛋白.方法:广西眼镜蛇毒经SephadexG75凝胶、CM Sepharose CL-6B离子交换层析及FPLC mono-S柱层析分离纯化出Natrin蛋白.结果:从广西眼镜蛇蛇毒中分离纯化得到的Natrin蛋白,经SDS-PAGE电泳测定其分子量为25 ku,质谱测定其分子量为24.937 ku,测得其N端的序列为NVDFNSESTRRKKKQKEIVD,C端的序列为NIKK.结论:从广西眼镜蛇毒中分离出纯的Natrin蛋白组分.
