电针对大鼠脑缺血再灌注后早期功能恢复及VEGF表达的影响

         

摘要

Objective: To investigate the effect of electroacupuncture on VEGF in brain tissue after cerebral focal ischemia-reperfusion in rats. Methods: 48 male SD(Spraguer-dawley) rats were randomly divided into sham-operated group, model group and electroacupuncture group, and subjected to establish the model of middle cerebral artery occlusion and reperfusion (MCAO/R) by nylon monofilament suture. Electroacupuncture was performed in 3 hours after establishing of successful model, Electroacupucture group were needled acupoint of Neiguan,Sanyin-jiao,shuigou,baihui,30 minutes each time,one time a day. Sham operation group and model group were conventionally fed in cages,without any intervention. The rats in the 7 d, 14 d randomly selected 8,the neural function recovery was evaluated by Longa's score,and immunohistochemistry techniques were used to detect the expressions VEGF. Results: Neural function: the sham operation group rats had no neurological symptoms. Model group and electroacupuncture group,there were no different about neural function score at the 7 th day. On the 14 th day,neural functional recovery of electroacupuncture group rats was significantly better than that of the model group. Immunohistochemistry: Sham operation group rats have no VEGF positive cells in section at each time. VEGF positive cells appear around rats' cerebral ischemia 7 d after cerebral infarction. It decreased 14d after cerebral infarction. The difference was significant compared with sham operation group. VEGF positive cells of electroacupuncture group increased significantly compared with the model group at each time. Conclusion: Electroacupuncture can effectively improve neurological function of cerebral infarction rats. The function enhancement may be partly due to the up-regulation of expression of VEGF expression in peri-infarct regions.%目的 探讨电针对脑局灶性缺血再灌注模型大鼠早期神经功能恢复和脑组织血管内皮细胞生长因子(VEGF)表达的影响.方法 健康成年雄性SD大鼠随机分为假手术组、模型组和电针组,每组16只.采用线栓法制作大鼠大脑中动脉缺血再灌注模型.电针刺激在造模成功90 min内进行,电针组针刺双侧内关、水沟、三阴交及百会,留针30 min,每日针刺1次.假手术组和模型组不进行任何干预治疗.各组大鼠在7、14 d两个时间点取8只进行神经功能评估及免疫组化SP法观察VEGF的表达.结果 假手术组大鼠无神经功能缺损症状.7d时模型组和电针组神经功能评分比较无明显差异.14 d时,电针组大鼠神经功能恢复明显优于模型组.假手术组可见少量VEGF的表达,模型组大鼠脑梗死后7d,模型组脑缺血周围出现VEGF表达增多,14 d持续增加,与假手术组比较均有明显差异.电针组VEGF表达在各时间点较模型组显著增加.结论 电针能通过促进脑局灶性缺血再灌注大鼠脑组织内VEGF的表达,有效改善大鼠局灶性脑梗死后的神经功能.
