不同培养基对山慈姑种子无菌萌发的影响

     

摘要

[目的]寻找山慈菇(Pleione bulbooioides(Franch.)Rolfe)种子萌发的最佳培养条件,为山慈菇试管苗的获得提供技术支撑.[方法]人工授粉获得山慈姑种子,利用其未成熟胚进行种子萌发试验,探讨了不同蔗糖浓度、不同活性炭浓度、不同类型的基本培养基、4种单激素(6 - BA、KT、NAA、IAA)及各自不同浓度、2种双激素组合及不同浓度等对山慈姑种子萌发率的影响.[结果]诱导山慈姑种子萌发的最适培养基为1/2MS+1.5 mg/L IAA+0.05%干酪素+2%蔗糖+0.75%活性炭+0.55%琼脂(pH 5.8),且添加了IAA的处理萌发率普遍较高,不仅诱导出原始球茎数量多,而且体积大,平均直径达1~1.5 mm.[结论]该试验建立优质、高效的山慈菇种子萌发体系,可为大量试管苗的获得奠定基础.%[Objective] To discover the optimum culture condition for seed germination of Pleione bulbocodioides (Franch. ) Rolfe and provide technical support for the acquisition of test - tube plantlets. [ Method] Effects of sucrose concentrations,active carbon concentrations,basal medi um types ,4 hormones (6 - BA,KT,NAA and IAA) concentration and 2 combinations of double -hormones on the germination of Pleione bulboco dioides ( Franch. ) Rolfe seeds that were obtained through artificial pollination were investigated in immature embryos. [ Result] The optimal cul ture condition for seed germination was a combination of 1/2MS,2% sucrose ,0.05% casein ,0.75% active carbon ,0.55% agar and 1.5 mg/L IAA (pH 5.8). The addictive IAA could cause a higher germination frequency,which induced more and larger protocorms whose average diame ter reacheed 1-1.5 mm. [Conclusion] A good and effective seed germination system was established in this study,which would provide basis for acquisition of abundant test - tube plantlets of Pleione bulbocodioides ( Franch. ) Rolfe.
