卡那霉素和潮霉素对芦竹不定芽诱导与生长的影响

         

摘要

[ Objective ] The sensitivity of Arundo donax Linn. to kancymycin and hygromycin under tissue culture were studied in this research, which provided basis for the construction of Arundo donax Linn. gene project construction and screening of transgenic plants. [ Method] Explants of Arundo donax Linn. were cultured for three days and then transferred to adventitious buds induction and growth medium adding with different concentration of kancymycin and hygromycin. [ Result] The result showed that both of the two kinds of antibiotics had influences on adventitious buds induction and growth. In 5 - 50 mg/L hygromycin medium, both of the induction rates and growth of adventitious bud were restrained. Hygromycin caused the death of explants in 50 - 100 mg/L concentration. For kanamycin, the concentration of 5 -50 mg/L can improve the induction rate of adventitious bud. The results showed that hygromycin was prefect for transformation of Arundo donax linn.[ Conclusion] Hygromycin restrained the induction and growth of adventitious bud under low concentration, and was suitable for the mark of transgenic choice of Arundo donax Linn.%[目的]研究组培条件下芦竹(Arundo donax Linn.)对植物基因工程常用抗生素卡那霉素和潮霉素的敏感性,为芦竹基因工程载体构建及转基因植株筛选奠定基础.[方法]芦竹外植体预培养3 d后接种于添加不同浓度卡那霉素或潮霉素的不定芽诱导培养基中培养,研究2种抗生素对芦竹外植体不定芽诱导及生长的影响.[结果]2种抗生素在一定浓度下对芦竹外植体不定芽诱导和生长都有影响.潮霉素浓度在5~50 mg/L时,随着浓度增大,对芦竹不定芽诱导及生长的抑制作用均增强;浓度达到50~100 mg/L时,外植体生长、分化逐渐停止直至死亡.卡那霉素浓度在5~100 mg/L时,对不定芽诱导产生促进作用.[结论]潮霉素在较低浓度下对芦竹外植体不定芽诱导和生长产生抑制作用,适合作为芦竹转基因选择标记.
