纤溶酶原激活物抑制剂1在肝纤维化组织中的表达及意义

摘要

目的 探讨纤溶酶原激活物抑制剂1(PAI-1)与肝纤维化发生的关系.方法 对30例乙型肝炎患者行肝脏活检,明确肝纤维化分级.正常肝组织3例,1～2级肝纤维化12例,3级肝纤维化8例,肝硬化7例.采用免疫组化及半定量分析方法,检测PAI-1在肝纤维化组织中的表达.结果 在正常肝脏中,PAI-1仅在汇管区细胞浆内有少量表达;肝纤维化组PAI-1表达均高于正常肝组织(P<0.05),且随纤维化程度而加重(P<0.05).在纤维化肝脏,PAI-1主要分布于细胞浆.结论 PAI-1在肝纤维化组织中高表达可能在肝纤维化的发生中起重要作用.%Objective To investigate the relationship and significance between expression of plasminogen activator inhibitor-l(PAI-1) and hepatic fibrosis. Methods On the basis of liver biopsy,20 patients with hepatitis B were classified as normal liver tissues in 3 cases, hepatic fibrosis grade 1 to 2 in 12 cases,grade 3 in 8 cases,and liver cirrhosis in 7 cases. PAI-1 expression in the liver tissues was detected by immunohistochemistry and semi-quantitative method. Results There was only weak expression of PAI-1 in normal liver. The expression of PAI-1 was significantly higher in the tissues with hepatic fibrosis(P＜0.05), which increased accordingly with severity of fibrosis(P＜0. 05) and mainly located within endochylema. Conclusion Overexpression of PAI-1 in fibrotic liver may play an important role in the pathogenesis of hepatic fibrosis.
