胃癌组织中细胞周期素D1的表达与细胞周期的关系

摘要

Objective To study the expression of cyclin Dl in gastric cancer and its relationship with cell cycle. Methods Western blot and flow cytometry techniques were applied to detect the expression of cyclin Dl and cell cycle distribution in cancer tissues(group A) and the tumor-adjacent normal gastric mucosa(group B) of 33 cases with gastric cancer. Results The positive expression rate of cyclin Dl was higher in group A than that in group B(63. 6% vs. 21. 1%) (F<0. 01). The cell ratio of G1/G0 phase was lower,but that of S phase was higher, in the cyclin D1-positive cases than that in the cyclin D1-negative in group A[(42. 2+1. 5)% vs. (64. 3±4. 8)% and (55. 1 + 1. 8)% vs. (34. 5 + 4. 9)%](P<0. 01). Conclusion The overexpression of cyclin Dl predicts a high activity of cell proliferation,which may take an important role in the carcinogenesis and development of gastric cancer.%目的 研究胃癌组织中细胞周期素D1( cyclin Dl)的表达,并分析其与细胞周期的关系.方法 采用Western blot和流式细胞术检测33例胃癌组织及其相应正常胃黏膜周期素D1表达和细胞周期分布.结果 胃癌组织周期素D1阳性率63.6％,高于正常胃黏膜的21.1％(P＜0.01).周期素D1阳性组胃癌G1/G0期细胞比例为(42.2±1.5)％,显著低于周期素D1阴性组的(64.3±4.8)％(P＜0.01);周期素D1阳性组胃癌S期细胞比例为(55.1±1.8)％,显著高于周期素D1阴性组的(34.5±4.9)％(P＜0.01).结论 胃癌细胞高表达细胞周期素D1预示癌细胞具有高增殖活性.周期素D1在胃癌发生、发展过程中可能起着重要的作用.
