目的:探讨杜仲提取物(EE)抗UVA致人真皮成纤维细胞光老化的作用机制.方法:将体外培养的ESF-1细胞通过UVA照射建立细胞光老化模型,给予500μg/mL、250μg/mL、125μg/mL、62.5 μg/mL4种不同浓度的杜仲提取物,采用ELISA法检测细胞培养液中Ⅰ型胶原蛋白(Col Ⅰ)、基质金属蛋白酶-1(MMP-1)、基质金属蛋白酶-3(MMP-3)的含量.结果:ESF-1细胞接受40J/cm的UVA照射后,细胞培养液中MMP-1、MMP-3含量明显升高(P<0.05),ColⅠ含量明显降低(P<0.05);给予EE后,细胞培养液中Col Ⅰ含量明显升高(P<0.05),MMP-1含量明显降低(P<0.05),MMP-3含量无明显变化.结论:杜仲提取物能抑制UVA引起的人MMP-1及MMP-3的异常分泌,降低Col Ⅰ的降解.%To investigate the mechanism of Eucommia extract against photoaging of human fibroblasts induced by UVA radiation. Methods: Cultured normal skin fibroblast was irradiated with UVA to develop photoaging model and treated with different dose of Eucommia extract. The amount of MMP - 1 、MMP - 3、Col I was detected by ELISA. Results; Cultured normal skin fibroblast irradiated by 40J·cm-2 , the amount of MMP - 1 、MMP - 3 increased obviously ( P < 0. 05 ) , Col I decreased obviously (P < 0. 05 ) ; after provided with Eucommia extract, the amount of MMP - 1 decreased obviously(P <0. 05) , the amount of Col I increased obviously(P <0. 05) and MMP - 3 was not obvious. Conclusions; Eucommia extract could inhibite the level of MMP -1 and MMP - 3 and reduce the degradation of Col I that induced by UVA.
