头孢曲松钠抗慢性神经病理性痛的作用机制研究

         

摘要

Objective To observe the effects of ceftriaxone ( Cef) on chronic neuropathic pain after sciatic nerve ligation ( CCI) .Method s Sixty male Spague-Dawley rats were randomly divided into sham-operation group;CCI group;Cef prevention group;Cef treatment group, at the same time, the rats who were injected intraperitoneally with NS were used as control group,with 10 rats in each group.Thermal withdrawal latency and mechanical withdrawal threshold were measured at 1, 3,5,7, 9,11 and 14 days after operation.Resutl s The CCI induced thermal hyperalgesia and mechanical allodynia in rats.In Cef prevention group, the thermal withdrawal latency at 5, 7 days and mechanical withdrawal threshold in the hindpaw ipsilateral to CCI at 7 days after operation were obviously prolonged,as compared with those in NS control group and CCI group ( P <0.05).In Cef treatment group,the thermal withdrawal latency at 11,14 days after operation,and mechanical withdrawal threshold in the hindpaw ipsilateral to CCI at 14 days after operation were significantly prolonged,as compared with those in NS control group and CCI group( P <0.05).Conclusion Cef has preventive and therapeutic effects on chronic neuropathic pain.%目的:观察β-内酰胺类抗生素头孢曲松钠( Ceftriaxone,Cef)对慢性神经病理性痛过敏的影响。方法雄性SD大鼠60只,随机分为Sham组、CCI(坐骨神经结扎)组、Cef预防组、Cef治疗组,后2组设腹腔注射0.9%氯化钠溶液组作为对照,每组10只。于术后第1天、第3天、第5天、第7天、第9天、第11天和第14天测定热缩足潜伏期和机械缩足反射阈值。结果 CCI诱导大鼠产生了热和机械性痛敏,Cef预防组大鼠CCI侧后肢热缩足反射潜伏期于CCI后第5天、第7天,机械缩足反射阈值于术后第7天同NS对照组以及CCI组相比明显延长( P <0.05);Cef 治疗组大鼠CCI侧后肢热缩足反射潜伏期在术后第11天、第14天,机械缩足反射阈值于术后第14天同NS 对照组以及CCI组相比明显延长( P <0.05)。结论 Cef对慢性神经病理性痛起到了预防和治疗作用。
