按酵母基因优化的HPV6型L1蛋白在大肠杆菌中高效表达

         

摘要

Human papillomavirus 6(HPV6) causes a common sexually transmitted disease,the genital warts.L1-capsid protein is a highly promising preventive vaccine that has entered phase Ⅱ clinical trial.But the existing methodologies for producing L1-capsid protein in insect cells is expensive for use in developing countries.In order to get larger amount of L1 protein in E.coli economically,we rebuilt whole L1 gene with preferred codons for yeast P.pastoris,abolished A-T rich regions,subcloned the modified L1 gene (mL1) into pTXB1 and induced expression by IPTG.SDS-PAGE and Western blot showed that mL1-6×His fused proteins could be overexpressed in BL21(DE3).This success will facilitate the HPV vaccine development and structure-function study.
