超声联合反义c-myc寡核苷酸对移植静脉内膜增生的影响

     

摘要

Objective To assess the effect of ultrasound conjunction with antisense c-myc oligodeoxyuncleotides treatment on intimal hyperplasia in a vein autograft model which is to find the new method in prevention of intimal hyperplasia.Methods 24 rabbits were divided into 4 groups randomly, which inciuding of control group, ultrasound group, antisense c-myc group and ultrasound conjunction with antisense c-myc oligodeaxynucleotides group,and 6 rabbits in each group.Fabricating a common carotid ar tery interpo sition model with ipsilateral internal jugular vein.The vein grafts treated by ultrasound (2.190 w/cm2 ,0.8 MHz) and ultrasound conjunction with antisence c-myc oligodeoxynucleotides.Neointimal thickness was calculated by histomorphometric quantification,expression of PCNA by immunostainning and c-myc expression by RT-PCR.Results Compared with the control group,after the fourth week the neointimal thickness,the number of PCNA positive cells and c-myc expression in ultrasound,antisense and ultrasound +antisense other groups were decreased significantly (P<0.01).Conclusion Treatment with ultrasound and antisense c-myc oligodeoxynucleotides seemed to decrease intimal hyperplasia and decelerate SMC proliferation.Therefore, ultrasound conjunction with antisense c-myc oligodeoxyuncleotides might be an effective treatment to reduce vein graft restenosis.%目的 观察超声辐照联合局部应用反义c-myc寡核苷酸对移植静脉内膜增生的影响,以期寻找防治内膜增生的新方法.方法 将24只兔随机分为4组,对照组、超声辐照组、反义核苷酸组、超声辐照加反义核苷酸组,每组6只,建立兔自体颈内静脉移植替代颈总动脉模型.超声辐照组术后当天至第7天使用超声(2.190 W/cm2,0.8 MHz)辐照移植血管段,反义核苷酸组局部转染反义c-myc寡核苷酸.观察移植静脉的组织形态学改变;增殖细胞核抗原(PCNA);反转录PCR(RT-PCR)检测移植血管段c-myc基因的表达.结果 术后第4周超声辐照组、反义核苷酸组、超声辐照加反义核苷酸组内膜厚度、PCNA阳性细胞数、c-myc基因表达均明显低于对照组(P<0.01).结论 超声辐照和局部转染反义c-myc可以降低平滑肌细胞的增殖和减轻内膜增生,可作为静脉移植后再狭窄的防治方法.
