目的 研究细胞粘附分子E-钙粘素(E-cadherin,E-cad)、γ-连环素(γ-catenin,γ-cat)及基质金属蛋白酶-9(matrix metalloproteinase9,MMP-9)与原发性肝癌侵袭转移的关系.方法 采用免疫组织化学方法,对经过石蜡包埋的40例原发性肝细胞癌组织和40例癌旁正常肝组织标本进行检测,并结合相关临床病理指标进行半定量分析.结果 E -cad、γ -cat及MMP-9在肝癌中的阳性表达率分别为30.0％ (12/40)、32.5％ (13/40)、77.5％ (31/40),在癌旁正常肝组织中的阳性表达率分别为100％ (40/40)、100％( 40/40)、17.5％ (7/40).E-cad、γ-cat在肝癌组织中不同程度的表达减弱或缺失,在癌旁正常肝组织中全部正常表达,MMP-9在肝癌组织呈现高表达,在癌旁正常肝组织中呈现低表达或不表达,三者的表达随肝癌的侵袭转移而有显著性差异(P＜0.05),且E-cad、γ-cat在肝癌组织中的表达都与MMP-9的表达水平呈负相关(r=-0.562,P＜0.05；r= -0.649,P＜0.05).结论 E-cad、γ-cat 及MMP-9参与肝癌的侵袭转移过程,在原发性肝癌恶性进展方面起着重要作用.%Objective To investigate the correlation of expression of E-Cad, γ-cat and MMP-9 with invasion and metastasis in human hepatocellular carcinoma ( HCC ). Methods The expression of E-cad, γ-cat and MMP-9 were detected immunohistochemically in 40 cases of HCC and 40 adjacent normal hepatic tissues. The results were densitomet-rically semiquantitated combined with related clinical pathological indicators. Results The positive expression rates of E-cad, γ-cat and MMP-9 were 30.0% (12/40) , 32.5% (13/40) , 77.5% (31/40) in HCC respectively, but 100% (40/40) ,100% (40/40) , 17.5% (7/40) in the adjacent normal hepatic tissues respectively. The expression of E-cad and γ-cat reduced or were negative in HCC, but E-cad and -y-cat expressed normally in all adjacent normal hepatic tissues. MMP-9 showed high expression in HCC, low or no expression in the adjacent normal hepatic tissues. The expression of E-cad,γ-cat and MMP-9 had significant difference (P ＜0.05) with invasion and metastasis of HCC. Furher-more, the expression of E-cad, "y-cat respectively were related with the expression of MMP-9 (r = -0. 562,P ＜0. 05; r=-0. 649,P ＜0.05). Conclusion The expression of E-cad, γ-cat and MMP-9 involves in the process of invasion and metastasis, play important roles in malignancy progress of HCC.
