环境设置对局灶性脑梗死大鼠梗死灶周围神经丝蛋白、突触膜糖蛋白表达的影响

         

摘要

目的 观察学习环境对局灶性脑梗死大鼠神经丝蛋白(NF)、突触膜糖蛋白(Syn)表达的影响.方法 SD大鼠110只,随机分为假手术对照组(对照组,10只)和大脑中动脉阻塞(MCAO)组(100只).MCAO组大鼠采用电凝法造成右侧MCAO模型后,再随机分为独居亚组(30只)、社会交往亚组(社交亚组,40只)和探索学习亚组(探索亚组,30只).于术后7d、14d、21d、28d分批处死大鼠,用免疫组化染色观察梗死灶周围皮质NF、Syn阳性表达.结果 MACO后7d不同环境设置亚组动物NF的表达水平均低于对照组(P<0.05),但其表达逐渐增高,28d时,探索亚组、社交亚组高于对照组(P<0.05);MCAO后7d、28d不同环境设置亚组梗死灶周围皮质Syn阳性表达均高于对照组(P<0.05);探索亚组于MCAO术后各时间点梗死灶周围皮质NF、Syn表达均明显优于独居亚组和社交亚组(P<0.05,P<0.01);第7天、21天、28天社交亚组NF表达均优于独居亚组(P<0.05),第7天、14天、21天社交亚组Syn表达均优于独居亚组(P<0.05).结论 探索学习环境、社会交往环境能促进梗死灶周围NF、Syn的表达.%Objective To observe effects of different environmental intervention on neurofilamen< NF ) and synapto-physin( Syn Expression in rats after unilateral local cerebral infarction. Methods 110 SD rats were randomly divided into sham( n = 10 ) and middle cerebral artery occlusion ( MCAO ) groups( n = 100 ). The rats in MCAO group were subjected to electric coagulation of MCA and then were assigned randomly to three subgroup, individual living subgroup( n =30 ), social communication subgroup( n =40 ), learning subgroup( n =30 ). The rats were randomly sacrificed at different time points( 7 d,14 d,21 d,28 d after operation ). The expression of NF and Syn in peri-ischemic cortex was examined by immunohistochemistry staining. Results After MCAO, the expression of NF and Syn in learning subgroup was higher than other subgroups at any time poin< P < 0. 05, P < 0. 01 ). In addition, the expression of NF in social communication subgroup was stronger that in individual living subgroup on day7, 21 and 28( P <0.05 ),the expression of Syn in social communication subgroup was stronger that in individual living subgroup on day 7, 14 and 2l( P < 0. 05 ). Conclusion Learning and social communication environment after MCAO could up-regulate the expression of NF and Syn in the peri-ischemic regions.
