活化的T细胞核因子信号通路在新疆哈萨克族高血压患者中的表达及其药物干预的试验研究

摘要

objectiveTo study the expression of nuclear factor of activated T lymphocytes C1 (NFATC1) and its downstream molecules' change before and after intervention in peripheral blood Tlymphocytes in Xinjiang Kazakh patients with essential hypertension (EH).MethodsThe 40 untreated Kazakh hypertensive patients and 20 Kazakh healthy subjects from Xinjiang were selected randomly in this study. The 40 untreated subjects were divided into 2 groups randomly: VIVIT group and hypertensive group, T-lymphocytes were isolated from peripheral blood with magnetic cell sorting and cultured for 120 h. mRNA and protein's expression level in 3 groups were detected by the technique of quantitive reverse transcription polymerase chain reaction and Western blot analysis respectively. One way ANOVA was taken in the comparision of variables data among 3 groups.ResuItsThe mRNA levels of NFATC1 in hypertensive group, VIVIT group and controls were 6.08±0.66, 3.84±0.22, 1.04±0.26, respectively (P<0.05), the mRNA levels of TNF-α in hypertensive group, VIVIT group and controls were 11.9±3.01, 2.26±0.27, 1.12±0.66, respectively. Contrast to VIVIT group and controls, the mRNA expression of TNF-αwere significantly increased in hypertensive group (P<0.05), meanwhile, the mRNA expression of TNF-αwere slightly higher than controls (P>0.05). The protein levels of NFATC1 in hypertensive group, VIVIT group and controls were 0.39±0.03, 0.14±0.02, 0.12±0.05, respectively. Contrast to VIVIT group and controls, the protein expression of NFATC1 were significantly increased in hypertensive group (P<0.05), meanwhile, the protein expression of NFATC1 were slightly higher than controls (P>0.05). The protein levels of TNF-α in hypertensive group, VIVIT group and controls were 0.44±0.06, 0.21±0.03, 0.25±0.02, respectively. Contrast to VIVIT group and controls, the protein expression of TNF-α were significantly increased in hypertensive group (P<0.05), meanwhile, the protein expression of TNF-α were slightly lower than controls (P>0.05).ConcIusionNFAT signaling pathways activate and participate in the pathological process of inflammation in essential hypertension and play an important role in Xinjiang Kazakh patients with essential hypertension. The inhibitor of the signaling pathways can reduce the production of inflammatory cytokines in the etiology and development of EH.%目的:观察新疆哈萨克族高血压患者外周血T淋巴细胞钙调磷酸酶/活化的T细胞核因子(CaN/NFAT)信号通路是否激活,并观察 VIVIT 干预前后炎症因子的表达情况。方法通过读取随机数字表的方法随机选取40例首次就诊且未经药物治疗的新疆哈萨克族高血压病患者,其中20例作为高血压组,20例为VIVIT药物干预组,同时选取20例哈萨克族健康体检者作为对照组。每例入选者均经肘静脉采血10 ml,密度梯度离心法分离出单个核细胞,免疫磁珠法分离三组患者外周血T淋巴细胞后增殖培养120 h,其中干预组采用活化的T细胞核因子阻滞剂VIVIT 2μmol/L进行干预。反转录聚合酶链反应法(RT-PCR)检测三组间mRNA的拷贝数,Western blot法检测对应蛋白的表达量,计量数据多组间的比较采用重复测量方差分析。结果 NFATC1在高血压组、VIVIT组、对照组的mRNA表达量分别为6.08±0.66,3.84±0.22,1.04±0.26,P均<0.05;TNF-α在高血压组、VIVIT组、对照组的mRNA表达量分别为11.9±3.01、2.26±0.27、1.12±0.66,分别与对照组、VIVIT组相比,高血压组TNF-α的表达量明显升高,P均<0.05,而VIVIT组TNF-α的表达量稍高于对照组,P>0.05;NFATC1在高血压组、VIVIT组、对照组的蛋白表达量分别为0.39±0.03,0.14±0.02,0.12±0.05,分别与对照组、VIVIT组相比,高血压组NFATC1的蛋白表达量明显升高, P均<0.05,与对照组相比,VIVIT组的NFATC1蛋白表达量稍高于对照组,P>0.05;TNF-α在高血压组、VIVIT 组、对照组的蛋白表达量分别为0.44±0.06,0.21±0.03,0.25±0.02,分别与对照组、VIVIT组相比,高血压组TNF-α的蛋白表达量明显升高,P均<0.05;而VIVIT组TNF-α的蛋白表达量稍低于对照组,P>0.05。结论新疆哈萨克族高血压患者外周血 T 淋巴细胞中存在CaN/NFAT信号通路的激活,从而参与高血压的低级别炎症的病理过程,CaN/NFAT信号通路抑制剂VIVIT可有效减弱经CaN/NFAT信号通路的炎性因子的生成。
