HPLC-UV法测定给药小鼠脑组织中5-羟色胺含量

             

摘要

Objective To establish a HPLC -UV method for determination of 5 - hydroxytryptamine (5 -HT) contents in administrated mice cerebral tissue. Methods The method was established with metaraminol as the internal standard (IS) ,the 280 run as the detection wavelength and 1 mL · min-1 as the flow rate. The mobile phase is aqueous phase (EDTA 0. 20 g and NaAC 2. 00 g were dissolved in 1000 mL water) - methanol -acetonitrile (500:40:20), phosphoric acid adjust pH 6.40. Results The linear range of calibration curves were 0. 26 - 42. 38 nmol · g-1. Precision RSD was 1.23%. Stability RSD was 2.40%. Conclusion This method could be used for determination of administrated mice cerebral tissue.%目的 用HPLC-UV法测定给药小鼠脑组织中的5-羟色胺(5-HT)含量.方法 以水相(EDTA 0.20 g,无水乙酸钠2.00 g,溶解于纯净水1000 mL中)-甲醇-乙腈(500:40:20),磷酸调节pH值至6.40为流动相,检测波长:280 nm,流量:1.0 mL·min-1.结果 在0.26~42.38 nmol·g-1内,线性关系良好;精密度RSD为1.23%;稳定性RSD为2.40%.结论 用HPLC-UV法测定小鼠脑中5-HT含量,方法可行.
