目的:探讨单核细胞在体外转分化为淋巴管内皮细胞的可能性.方法:取成人新鲜外周血,分离单核细胞,分别在层粘连蛋白(FN)铺板或血管内皮生长因子-C(VEGF-C)、白介素-3(IL-3)、白介素-7(IL-7)刺激下诱导培养24 h,通过形态学分析、RT-PCR和免疫荧光细胞化学分析方法检测单核细胞中淋巴管内皮特异性标志物LYVE-1、podoplanin、Proxl及内皮细胞共同抗原vWF、eNOS的表达.在此基础上,进一步用内皮细胞培养基-2(EGM-2)、VEGF-C和FN铺板联合诱导单核细胞向淋巴管内皮细胞转分化.结果:单核细胞经FN、VEGF-C、IL-3、IL-7短暂诱导即能表达LYVE-1、podopla-nin、Proxl,而不表达vWF、eNOS.结论:单核细胞在炎症或者内皮细胞生长环境中可以表达淋巴管内皮细胞标志物,但尚不能证实转化为真正的淋巴管内皮细胞.%Objective: It has been recently confirmed that monocytes can differentiate into endothelial cells, and therefore, attribute to angiogenesis. However, whether the monocytes can transdifferentiate into lymphatic endothelial cells is still unknown. Methods: Fresh monocytes from peripheral blood were collected and then incubated in FN-plated well or treated with VEGF-C, IL-3, IL-7 for 24 hours or 6 days. The expression of specific markers of lymphatic endothelial cells such as LYVE-1, Podoplanin and Proxl, and the common markers of endothelial cells such as vWF and eNOS were detected by morphology. RT-PCR and immunochemical methods. After the induction, we further induced the monocytes to express the lymphatic endothelial markers by EGM-2, VEGF-C and FN. Results: The experiments revealed that monocytes could express lymphatic endothelial markers LYVE-1, Podoplanin and Prox-1 briefly, but not endothelial markers vWF and eNOS. Conclusion: Monocytes may have the ability to express the lymphatic endothelial markers in the inflammation conditions or endothelial growth environment Nevertheless, we could not confirm the cells to transdifferentiate into actual lymphatic endothelial cells.
