人参锈腐生防菌Psn86的GFP标记及在土壤和有机肥中的定殖示踪

         

摘要

为跟踪分析从人参内生菌哈茨木霉(Trichoderma harzianum)中优选的人参锈腐生防菌Psn86在有机肥及土壤中的存活能力与定殖特性,利用农杆菌介导的真菌遗传转化体系,获得了含有绿色荧光蛋白基因(gfp)的哈茨木霉标记菌株Psn86-g.结果表明,该菌株在生长速率、产孢量、对酸碱度和温度的适应性及对人参锈腐菌的抑菌活性等方面与原始哈茨木霉菌株Psn86无显著差异(P>0.05).同时研究了标记菌株Psn86-g在有机肥与山林土混合物、有机肥、山林土3种处理中的定殖能力以及人参生长对该菌株在3种处理中定殖的影响.结果显示,标记菌株Psn86-g在土肥混合的处理组中定殖中最好,其次为山林土中,在有机肥中定殖最差.种植人参比未种植人参更有利于菌株的存活.标记菌株Psn86-g在不同处理组中的菌落数随时间的延长均略有下降,16 d后趋于稳定,维持在1×105cfu/g,较初始接菌量下降了约1个数量级.%Psn86 was a biocontrol strain which was selected fro m ginseng endophytic fungia Trichoderma harzianum. In order to study survival and colonization characteristics of Psn86 in organic manure and soil, T. harzianum strain marked with green fluorescent protein (GFP)gene named Psn86-g was obtained by Agrobacterium mediated fungal genetic transformation system.The results showed that there were no significant difference between Psn86 and Psn86-g(P>0.05)in growing rate, quantity of sporulation, adaptability of pH and temperature and inhibition activity against ginseng Cylindrocarpon destructans.Capacity of colonization of Psn86-g in organic manure,soil and mixture of organic manure and soil were studied.The results showed that Psn86-g colonization capacity was the best in mixture treatment,followed by soil, and that of organic fertilizer treatment was the worst. Condition of planting ginseng was more favorable for survival of Psn86-g. Colony number of Psn86-g decreased slightly with the extension of time in different treatment groups,tended to be stable after 16 days at 105CFU/g,which dropped by about 1 orders of magnitude comparing with the initial inoculum size.
