不同剂量ALA-PDT对皮肤鳞状细胞癌A431细胞增殖的抑制作用

         

摘要

目的:确定5-氨基酮戊酸-光动力疗法对皮肤鳞状细胞癌A431细胞抑制的最佳剂量.方法:体外培养人皮肤鳞状细胞癌A431细胞株,应用MTT法检测不同ALA浓度(0.5、1、2 mmol/L)、不同PDT光照剂量(5、10、20、40 J/cm2)下A431细胞的增殖水平.结果:当ALA浓度为0.5 mmol/L时,光照剂量为5、10、20、40 J/cm2对A431细胞的抑制率分别为6%、10%、15%、18%;当ALA浓度为1 mmol/L时,光照剂量为5、10、20、40 J/cm2对A431细胞的抑制率分别为30%、52%、80%、82%;当ALA浓度为2 mmol/L时,光照剂量为5、10、20、40 J/cm2对A431细胞的抑制率分别为31%、54%、81%、82%.结论:ALA在浓度为1 mmol/L,光照剂量为20 J/cm2时,ALA-PDT对A431细胞增殖抑制效果达到最佳.%Objective:To determine the optimal dose of ALA-PDT on the inhibition of A431 cells proliferation in cutaneous squamous cell carcinoma.Methods:A431 cells were cultured firstly in vitro, and then, were treated with different drug concentration (0.5, 1 and 2 mmol/L) and different dose of PDT (5, 10, 20 and 40 J/cm2).The proliferation of treated A431 cells was detected by MTT.Results:The inhibition rates of A431 cells treated under the condition of 0.5 mmol/L ALA combined with 5, 10, 20 and 40 J/cm2 PDT were 6%, 10%, 15% and 18% respectively.The inhibition rates of A431 cells treated with 1 mmol/L ALA combined with PDT of 5, 10, 20 and 40 J/cm2 PDT were 30%, 52%, 80% and 82% respectively.The inhibition rates of A431 cells treated with 2 mmol/L ALA combined with 5, 10, 20 and 40 J/cm2 PDT were 31%, 54%, 81% and 82% respectively.Conclusion:The highest inhibition rate of PDT on A431 cell proliferation is 1mmol/L ALA and 20 J/cm2 PDT.
