不同龋敏感儿童牙菌斑微生物多样性分析

     

摘要

目的:利用高通量测序方法,研究 ECC 儿童和 S-ECC 儿童牙菌斑微生物的多样性变化,探索儿童龋病的病因学。方法采集 ECC儿童和 S-ECC 儿童牙菌斑样本,利用 Il umina MiSeq 测序方法,针对16SrRNA 基因 V4变异区设计通用引物进行测序分析菌群微生物的群落变化。结果测序总共测得1669057条有效序列,平均有效率为89.16%,共1514个操作分类单位(OTU)。结论儿童龋病的发生发展与链球菌属等菌属密切相关,其种群的多样性呈现特异性的富集。%Objective To study the microbial diversity of ECC and S-ECC in the dental plaque with high-throughput sequencing, and investigate the possible etiology of dental caries of children. Methods Describe the bacterial profile shifting in different caries states by pyrosequencing analysis based on 16SrRNA gene V4 hyper variable regions. Apply Illumina MiSeq pyrosequencing for analysis random drawing for the ECC and S-ECC. Results In the clonal analysis, a total of 1 669 057 high-quality sequences were produced, accounting for 89.16 % of valid sequences, a total of 1 514 operating units (OTU). Conlusion With the occurrence and development of Streptococcus spp. and other dental caries in children is closely related to the diversity of the population showed specific enrichment.
