刺松藻多糖免疫调节与抗肿瘤增效研究

         

摘要

Objective To investigate the antitumor and immunomodulation activities of wide seaweed Codium fragile ( sur. ) Hariot poly-saccharides( CFPS) in vivo. Methods The antitumor effects of CFPS on Hca-F hepatoma-bearing mice were assessed by quality analysis and TUNEL assay. The lymphocyte antitumor-activity was assessed by MTT assays, the effect of CFPS on phagocytosis of macrophages was studied by neutral red method. The effect of CFPS on the level of cytokine secretion of the serum was detected by ELISA method. Results Compared with the control group, the antitumor activities obviously were improved( P < 0. 05 ); the antitumor rates in CFPS group,5-Fu group and CFPS plus 5-Fu group were 59.77% , 60.05% ,and 72. 84% respectively. There were signs of tumor-cell apopto-sis in CFPS group, 5-Fu group and CFPS plus 5-Fu group. A significant difference was observed in relative contents of IL-10, IL-12 and TNF-ot produced by CFPS in mice serum. Compared with the 5-Fu group, relative contents of IL-10, TNF-αproduced were higher in the CFPS plus 5-Fu group(P < 0.05 ). Conclusion CFPS may have great effects on hepatoma-bearing mice and act as a biological modulator, with antitumor activity. It can enhance the effect of anti-tumor efficacy of 5-Fu, and improve the prognosis.%目的 研究刺松藻多糖(CFPS)对荷瘤小鼠化疗的免疫调节与抗肿瘤增效.方法 以Hca-F肝癌小鼠为模型,采用重量分析法和TUNEL检测分析CFPS抗肿瘤作用;采用中性红比色法、MTT法检测CFPS对小鼠脾脏巨噬细胞吞噬活性、淋巴细胞杀瘤活性;ELISA法检测CFPS对血清中细胞因子分泌水平影响.结果 与对照组相比,CFPS组、5-Fu组和CFPS与5-Fu联合用药组抑瘤率分别为59.77%、60.05%,72.84%,抑瘤作用显著增强(P<0.05);CFPS组、5-Fu组、CFPS与5-Fu联合用药组肿瘤细胞出现明显凋亡迹象.CFPS调节细胞因子IL-10、IL-12、TNF-α分泌均有显著性作用(P<0.05),IL-10、TNF-α在5-Fu组、CFPS与5-Fu联合用药组间差异显著(P<0.05).结论 CFPS对荷瘤小鼠有免疫调节活性,在肿瘤的免疫治疗中发挥作用,能直接起到抑瘤作用.并可明显增强抗肿瘤药物5-fluorouracil(5-Fu)的抗肿瘤作用,可明显改善预后.
