针刺“百会”透“曲鬓”对实验性大鼠脑出血后灶周组织PAR-1和NF-κBp65表达的影响及其相关性分析

             

摘要

Objective: To study the dynamic expression and relativity analysis of PAR -1 and NF - κBp65 after intracere-bral hemorrhage (ICH) rats and the influence of acupuncture on them. Methods : The 96 healthy Wistar male rats were selected. 90 healthy male Wistar rats were randomly divided into 3 groups : model group , acupuncture group and west-em medicine group,30 rats in each group. Each group was examined at S different time points of 6 hours, lday, 2days, 3days and 7days, respectively, 6 rats in each group. Meanwhile,6 rats were taken as the blank control group. The in-tracerbral hematoma rat model was made by autologous blood injection by using stereotactic method. Western - blot analysis was used to observe the expression of Par - 1 and NF - KBp65 of the intracerebral hemorrage rats. Results: The expression of PAR -1 and NF -κBp65 had an obvious down - regulation at different phases in the brain of the acupuncture group rats, which showed significant differences compared with the model group (P < 0. 05 ). The correlation analysis was positively relative. Conclusion; The acupuncture of BaiHui - QuBin decreases the mechanism of inflammatory response after ICH to gain the antagonistic effect on brain edema and secondary brain injury by means of down - regulating the expression of NF - kBd65 which inhibits the transcription of NF -κBp65.%目的:探讨针刺对急性脑出血(Intracerebral hemorrage,ICH)后凝血酶受体-1(PAR-1)和核转录因子(NF-κBp65)动态表达的影响,并探讨两者的相关性。方法:选取健康雄性Wistar大鼠96只,将其中90只随机分为模型组、针刺组、茴拉西坦组(简称西药组)3组。每组又分为6h、1d、2d、3d和7d 5个时间点,每个时间点6只大鼠,另设空白对照组6只大鼠。制备ICH大鼠模型,采用Western-b1ot免疫印迹技术观察不同时间点头穴透刺治疗对ICH大鼠脑组织PAR-1、NF-κBp65蛋白表达的影响,并对两者进行相关性分析。结果:针刺组大鼠脑内PAR-1、NF-κBp65蛋白表达在不同时间点均有显著下调,与模型组比较PAR-1均有显著性差异(P<0.05),具有统计学意义。相关性分析两者呈正相关。结论:针刺百会透曲鬓穴可抑制ICH后凝血酶引起的毒性作用,进而下调NF-κBp65的表达,抑制了核转录因子-κB的炎症因子转录作用,达到减轻炎症级联反应程度,防治继发性脑损伤的作用。
