不同抗旱性柑橘砧木相关生理指标及基因表达差异分析

         

摘要

The objective of this study is to analyze the drought-resistance characteristics of different Citrus rootstocks so as to exploit drought-resistance resources of Citrus. Taking seedlings of ' Sanhu red tangerine' (Sanhuhongju) ( drought resistant) and ' Sour pummelo' (Citrus grandis ' sour pummelo' ) (drought sensitive ) as test materials, the discrepancy analysis on the physiological indexes such as plant height, relative water content, the activity of superoxide dismutase (SOD) , catalase (CAT) , peroxydase (POD) , the content of malonaldehyde (MDA) , proline, soluble sugar and the gene expression of sod, cat,pod,pro and lea was conducted. The results showed that 'Sanhu red tangerine' ,which was the drought resistant variety, mainly preserved water potential by absorbing the deeper soil water and it was more suitable to use plant height, relative water content, SOD, CAT and MDA as its drought - resistance evaluating indexes in ' Sanhu red tangerine'. The gene expression of sod,cat,pro and lea corresponded with the change of related physiological indexes. Their expression decreased under appropriate drought condition, but increased under drought stress. And compared with cat, sod might be the principal gene. However, the relation among genes still need further study.%为分析不同柑橘砧木抗旱特性,以期进行柑橘抗旱资源的开发,以三湖红橘(Sanhuhongju)(抗旱)和酸柚(Citrus grandis‘sour pummelo’)(干旱敏感)实生苗为材料,进行株高、叶片相对含水量、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、过氧化物酶(POD)、丙二醛(MDA)、脯氨酸、可溶性糖等生理指标及sod、cat、pod、pro、lea等基因表达量差异分析.结果表明,抗旱品种三湖红橘主要通过吸收深层土壤水分以维持水势,且株高、叶片相对含水量、SOD、CAT、MDA更适宜作为三湖红橘的抗旱评价指标.sod、cat、pro、lea基因表达量的变化与相应生理指标的变化相对应,在适宜生长的干旱条件下表达量下降,受胁迫危害时表达量上升,且相对于cat,sod可能为主效基因,但其基因间的具体关系还有待进一步研究.
