Skin is a major organ of the body that often needs repair. Autografts are the gold standard, however only a limited amount of tissue can be harvested from the body. In patients with serious skin burns or defects, a synthetic scaffold would be a more optimal means of tissue regeneration. The focus of the current study is on developing an electrospun scaffold that has a similar structure and function as native skin. To accomplish this goal, we have developed a unique electrospinning protocol that significantly increases porosity within nanofibers consisting of collagen type I and polycaprolactone (PCL). This method allows the generation of a composite scaffold consisting of both dermal and epidermal skin layers.
