PREPARATIVE CHROMATOGRAPHY AT SUPERCRITICAL CONDITIONS

The basic issues in packed bed supercritical fluid chromatography (SFC) are: ⅰ) characterizing adsorption equilibrium; ⅱ) characterizing mass transfer dynamics; ⅲ) predicting column dynamics and iv) the effect of polar modifiers. The first three issues have been addressed through experiments. Experiments have been performed on a preparative column with significant pressure drop using pure CO_2 as an eluent and phenanthrene dissolved in toluene as solute. Parameters relating to retention behaviour and mass transfer characteristics were measured. The observations are reported and the deviations with respect to HPLC and GC behaviour are highlighted and discussed.
