Preparation of the Chitosan-gelatin Sponge Wound Dressing and Its Bioactivities

By freezing the chitosan-gelatin compound with adding cross-linking reagent, and inducing phase abruption, the chitosan-gelatin sponge wound dressing (CGSWD) was made, which had unique porous membrane structure. Its structure, mechanical properties, moisture regain and retention were studied with many physical methods such as SEM, electron tension tester and so on. The antibacterial properties of the CGSWD dried by the way of natural dryness and microwave dryness were investigated, and then the antibacterial properties of the CGSWD were compared with that of cefradine, ciprofioxacin and penicillin. The animal wound models were made on the back of New Zealand rabbits, and then the wound were respectively treated with 0.2 % (v/v) ethacridine and CGSWD. The wound skin tissues were observed with Scanning Electron Microscope (SEM) at 14 days after initial wounding and compared with thenormal skin without any treatment. The results showed that: (1) The CGSWD possesses porosity, good permeability and moisture absorption property. (2) The CGSWD provides quite well antibacterial property. The antibacterial property of CGSWD by microwave dryness is better than that by natural dryness. By comparing the CGSWD antibacterial property to E.coli K88 and streptococcus with penicillin, ciprofioxacin and cefradine, their anti-E.coli K88 ability is as follows order: Cefradine, ciprofioxacin > CGSWD > Penicillin. The order of anti-streptococcus ability is like that: Antibioticses > CGSWD by natural dryness, Ciprofioxacin, penicillin > CGSWD by microwave dryness > Cefradine. (3) The effect of CGSWD exceeds 0.2 % (v/v) ethacridine in wound healing. Thus, the CGSWD is an excellent wound healing material.
