Polymer-based monolith materials with 3-dimensional interconnected porous structure have attracted attentions on various applications. In this study, a facile template-free method, namely thermally induced phase separation (TIPS), is proposed to prepare a monolith based on a reactive polymer, poly(glycidyl methacrylate-co-methyl methacrylate) (PGM). The protocol to prepare PGM monolith is as follows. PGM is dissolved in ethanol/H_2O by heating, followed by cooling the solution to induce the phase separation. After the solvent replacement with water and drying under vacuum, the monolith is obtained. The macroporous morphology of this PGM monolith can be readily controlled though the preparing conditions. Porous supports are widely used for enzyme immobilization to improve reusability. Aldehyde group is often used as a good functional group to immobilize an enzyme with involving reductive amination under mild conditions. The PGM monolith is modified with aminoacetal to yield an aldehyde group-bearing monolith (PGM-CHO), followed by immobilizing pepsin via reductive amination. The immobilized pepsin exhibited better pH stability and thermal stability compared to the free pepsin. Furthermore, this immobilized pepsin on the modified PGM monolith is applied to liquid chromatography-mass spectrometry (LC-MS) for protein online analysis.
