Identification and Characterization of A New Cucumber Mosaic Virus Isolate from Passionfruit in Hainan of China

A virus was isolated from Passiflora edulis f. flavicarpa grown in Hainan island of China. It was identified as a cucumber mosaic virus (CMV) on the basis of biological properties, particle morphology and serology, and confirmed by sequence comparison of coat protein (CP) gene. This new CMV isolate was named CMV-Pf. CMV-Pf could systemically induce mosaic and leaf curling of P. edulis. Biological reaction of this isolate on a number of major indicator plants by sap-inoculation was identical to that of the others CMV isolates, but difference was observed on Nicotiana tabacum var. Samsun. The CMV-Pf was purified and demonstrated to be isometric particles on electron microscope. Antiserum was produced by immunized rabbits with purified virus particles. Immuno double diffusion test and ELISA indicated that the virus isolate was serologically close related to CMV. SDS-PAGE analysis of CMV-Pf demonstrated that the virus contains a coat protein (CP) of 28 kD. The CP gene of CMV-Pf was cloned by means of RT-PCR method. Sequence analysis result indicated that the CMV-Pf is a member of subgroup I of CMV.
