iMALDI: (1) offers absolute quantitation of Ang-I from whole human plasma from linear standard curves with R~(2) values of 0.99. (2) has no cross-reactivity with Ang-II (absolute specificity is provided by MS and MS/MS analysis). (3) allows fast and highly accurate determination of PRA. (4) has advantages in time, safety and specificity in PRA determination compared to currently used assays (no use of radioisotopes, lengthy incubation times or liquid chromatography procedures). The agreement between iMALDI and RIA was assessed by comparing PRAs obtained from 49 different plasma samples. PRAs determined by iMALDI were found to be higher in all cases compared to those obtained by RIA; most probably due to the higher pH during Ang-I generation.
